Objective:To study of the primary comparison of quality of Podocrapus Nagi and Podocrapus Fleuryi Hickel, by applying the microphotography technology to study the pharmacognosy and finger printing technology was applied to the study of the comparison of flavonoids constituents and volatile constituents in Podocrapus nagi and Podocrapus Fleuryi Hickel, to reveal its characteristics of pharmacognosy and fingerprinting, to provide quality reference for the development and utilization of the two Podocrapus plants.Method:Microphotography and micro chemisty were applied to study the transverse section, surface spreading section and powder of Podocrapus Nagi and Podocrapus Fleuryi Hickel by description of oraganization structure and ergastic substance. TLC was applied to study the identification comparison of two medicinal materials. UV spectrophotometry was applied to determine the content of total flavonoids. HPLC was applied to determine the content of amentoflavone of two plants. The comparison of fingerprinting was appled to study the flavonoids constituents of Podocrapus Nagi and Podocrapus Fleuryi Hickel by HPLC. Volatile oils were obtained by steam distillation and were separated, analyzing and compared by GC-MS.Results:Powder of Podocrapus Nagi and Podocrapus Fleuryi Hickel showed that there was strong similarity in the microstructure and powders of leaves of two plants. But there was difference in that longer palisade tissue and greater amount of volatile oils in the leaves of Podocrapus Fleuryi Hickel. The result of TLC showed that there was mainly flavonoids constituents in Podocrapus Nagi and Podocrapus Fleuryi Hickel. There was great difference of flavonoids constituents in leaves, stem branch and skin of Podocrapus Nagi and Podocrapus Fleuryi Hickel. Those of leaves is the most , stem branch was next and skin was lowest.The method was established the determining the content of flavonoids constituents with the standard of lutin. The effect of extraction about total flavone from Podocrapus Nagi and Podocrapus Fleuryi Hickel was best with 50% ethanol.The content of amentoflavone was different in Podocrapus Nagi and Podocrapus Fleuryi Hickel. The range of content of amentoflavone in Podocrapus Nagi was from 0.1618 to 0.5721 mg/g. The range of content of amentoflavone in Podocrapus Fleuryi Hickel was from 0.039 to 0.1304 mg/g.Podocrapus Nagi and Podocrapus Fleuryi Hickel were analyzed by hplc fingerprint with almost total flavone, specially amentoflavone. The types of component were similarity in Podocrapus Nagi and Podocrapus Fleuryi Hickel, but the content were variant. Evaluating the similarity of the fingerprints showed that 8 batches Podocrapus Nagi had the high similarity of fingerprints with coefficient of above 0.9 and 5 batches Podocrapus Fleuryi Hickel had the high similarity of fingerprints with coefficient of above 0.9, too. Explicit classification between Podocrapus Nagi and Podocrapus Fleuryi Hickel was presence.18 peaks in volatile oil of leaves of Podocrapus Nagi were separated and 13 compounds was identified with 94.11 % total area of total ion chromatography peaks. 29 peaks in volatile oil of leaves of Podocrapus Fleuryi Hickel were separated and 13 compounds was identified with 91. 44% total area of total ion chromatography peaks. But there was great difference in the area of every common peak.Conclusion:The similarity of microscopic characteristics showed that Podocrapus Nagi and Podocrapus Fleuryi Hickel had a near relationship. But the difference that could be the microscopic identification basis was on the length of palisade cells of mesophyll tissue and the content of volatile oil in the leaves of Podocrapus Nagi and Podocrapus Fleuryi Hickel.It was first to study the tlc of Podocrapus Nagi and Podocrapus Fleuryi Hickel. The method of TLC and FeCl3 color was used for the differentiationof them.A method for determining total flavone was established with a simple, rapid, accurate and reproducibily characteristics.The content of amentoflavone in Podocrapus Nagi was singnificantly higher than that in Podocrapus Fleuryi Hickel. The content of amentoflavone could be difference characteristics and reference basis of quality evaluation of medicinal materials in Podocrapus Nagi and Podocrapus Fleuryi Hickel.The types of component were similarity in Podocrapus Nagi and Podocrapus Fleuryi Hickel, but the content were variant that was one difference characteristic of them especially for amentoflavone.Evaluating the similarity of the fingerprints showed that 8 batches podocrapus nagi had the high similarity and 5 batches Podocrapus Fleuryi Hickel had the high similarity,too. Explicit difference between podocrapus nagi and Podocrapus Fleuryi Hickel was used for identification characteristic and quality evaluation index.Terpenes and alcohols are major chemical constituents in the oil of Podocrapus Nagi and Podocrapus Fleuryi Hickel. There were similarities and differences in the compositions and analysis map in the oil by GC-MS. The method was simple, rapid and could be used for analysis and discrimination in eht oil of Podocrapus Nagi and Podocrapus Fleuryi Hickel.
