The study of this article was mainly on Tussilago farfara L.quality study,Tussilago farfara L.and The compound presciptions of Tussilago farfara L.and Aster tataticus L.f.extraction and purification process of total flavonoids and fingerprint;total saponins in Aster tataticus L.f.and fingerprint。In documentary study,it was concludend the study of the theory of Traditional Chinese Medicine of Tussilago farfara L.,Aster tataticus L.f.and The compound presciptions of Tussilago farfara L.and Aster tataticus L.f.,and Chemical Constituents and pharmacology study progess on Tussilago farfara L.and Aster tataticus L.f.In quality studies,the study established the criteria of quality control of Tussilago farfara L.,for it was not consummated in the former criterion which had only source and identification,the quality standard was incomplete which no content determination presented.The quality standard of Aster tataticus L.f.was comparative completely,which has content determination presented.The quality studies of Tussilago farfara L.,which concludes TLC qualitative test was conducted using lutin as reference components, confirmed the limits of content of extract.Determination of total flavonoids by ultra-voilet spectrophotometry and quantitative analysis by RP-HPLC were conducted to 10 samples from different habitats,using lutin as reference substance.The quality studies of Aster tataticus L.f., which concludes determination of total saponins by ultra-voilet spectrophotometry and quantitative analysis by HPLC-ELSD,using Shionone as reference substance.Studies were carried on the extraction and purification process of total flavonoids in Tussilago farfara L.and The compound presciptions of Tussilago farfara L.and Aster tataticus L.f..By reviewing the impact of ethanol concentration,ratio of herb to solvent,extraction time.Set up the L9(34)orthogonal test and confirmed the best extraction process of Tussilago farfara L.as 12BV 60%ethanol distilling three times,1h per time.And,as 10BV 80%ethanol distilling three times,30min per time,for The compound presciptions of Tussilago farfara L.and Aster tataticus L.f.Extract of total flavonoids was purified with macroeticular resins. Different kinds of resins were applied to review the static and dynamic adsorption ability,to optimize the concentration,volume and flow rate of eluant.It was confirmed that the best purification process of Tussilago farfara L.was XAD16 maeroeticular resins,total flavonoids of eluant to resins being 5:2,eluted by 20BV 40%ethanol after water with flow rate of 2ml/min.40%ethanol part was collected and dried under vacuum. 10 samples of Tussilago farfara L.resulting in the total flavonoids content more than 56%。And,as XAD16 macroeticular resins,eluted by 20BV 50%ethanol after water with flow rate of 2ml/min.50%ethanol part was collected and dried under vacuum resulting in the total flavonoids content more than 50%.Studies were carried on the extraction and purification process of total saponins in Aster tataticus L.f.Set up the L9(34)orthogonal test and confirmed the best extraction process as 8BV 95%ethanol distilling three times,1h per time.Extract of total saponins was purified with macroeticular resins.Different kinds of resins were applied to review the static and dynamic adsorption ability,to optimize the concentration, volume and flow rate of eluant.It was confirmed that the best purification process was XAD1 macroeticular resins,total saponins of eluant to resins being 1:2.6,eluted by 22.5BV 95%ethanol after water with flow rate of 2ml/min.95%ethanol part was collected and dried under vacuum.Studies of HPLC fingerprint was conducted to ten origins of Tussilago farfara L.and its total flavonoids extract(intermediate),Aster tataticus L.f.and The compound presciptions of Tussilago farfara L.and Aster tataticus L.f.Their fingerprints were built up by optimizing chromatogram condition,to characterize 15,17,21 and 18common peaks separately,so that the quality of medicinal materials can be controlled. Referring to "Technical Specifications of Fingerprint Study of Chinese Traditional Medicine Injection(Interim Regulation)" decreed by SFDA and similarity evaluation system for chromatographic fingerprint of TCM(2004 A)set by China pharmacopoeia committee,the results showed the similarity of Flos Farfarae and its total flavonoids extract(intermediate)were both from 0.9 to 1.0.Compare 18 common peaks areas among ten samples and the change of peak areas among Aster tataticus L.f.extracton with Tussilago farfara L of different ratio by academic peak areas.
