TP,tannic acid and gallic acid are the most extensive and typical polyphenols of human food. At present,these polyphenols have been proved to be have amultiple functional role on human beings,but the mechanism of polyphenols have not been clearly explained.As the foodborne polyphenols should be digest through gastrointestinal before absorbed,so most important premise is to study the polyphenols on the role of the human body through researching the relationship between polyphenols and digestive enzymes.The digestive tract habitats are complex,and polyphenols has a very lively chemical properties that it can be affected by acid,alkali,oxygen and other factors,under which conditions oxidate rapidly.The oxidation products can participate in a lot reaction because there chamical properties are lively.therefore,there is a significant disruption by many determination perticularly spectrophotometry,except special analysis conditions and reagent.This article explored a simple,accurate and well reproducible measurement of the activity of gastrointestinal protease,in order to study the relationship between the polyphenols and activity of digestive enzyme.Further more to explore the relationship between foodborne polyphenols,which take TP,tannic acid and gallic acid as the representative and the digestive tract protease activity such as trypsin and pepsin and the digestive tract activity of Lipase,such as pancreatic Lipase.The results shoued that:(1)Formaldehyde instructed by benchmark corrected acidimeter is a a reliable way to measure the content of free amino acid.Changing the test conditions,formaldehyde can be used to measure of trypsin and pepsin activity,the best determination conditions were as follows:pH 7.5,react 30 min, 37℃,the activity of trypsin was 9.10 U/mg,the Michaelis constant Km of trypsin's 1.4 mol/L using the double countdown mapping method.While the best determination conditions of the pepsin activity were as follows:pH 2.0,react 30 min,37℃,the activity of pepsin was 2.17 U/mg,the Km was 0.42 mol/L.(2)Formaldehyde instructed by benchmark corrected acidimeter is a a reliable and well reproducible way to measure the activity of Lipase,the best determination conditions were:pH 6.9, react 30 min,37℃,the activity of Lipase was 10.04 U/mg,the Km was 0.028mol/L. (3)TP can inhibit the activity of Trypsin,when added 1 mg/mL of TP 1mL,the relative activity fell to 53.7 percent,the inhibition rate up to 46.3%.Inhibit type was competitive inhibition,inhibition constant was 0.07 mg/mL;TA can also inhibit the Trypsin activity,when added 1 mg/mL of TA 1mL,the relative activity fell to 76.7 percent,its inhibition rate up to 23.3%,Inhibit type was competitive inhibition,inhibition constant was 0.09 mg/mL.GA also inhibit the activity of Trypsin, when the concentration of GA was 0.0053 mg/mL,the relative activity fell to 16.3 percent,the inhibition rate was 83.7%,the inhibit type was competitive inhibition,inhibition constant Ki was 1.03×10-4mg/mL.(4)TP can inhibit the activity of Pepsin,when added 1 mg/mL of TP 1mL,the relative activity fell to 61.2 percent,its inhibition rate up to 38.3%.Inhibit type was competitive inhibition,inhibition constant was 0.05 mg/mL;TA can also inhibit the Pepsin activity,when added 1 mg/mL of TA 1mL, the relative activity fell to 72.4 percent,its inhibition rate up to 27.6%,the inhibit type was competitive inhibition,inhibition constant was 0.012 mg/mL;GA also inhibit the activity of Pepsin, when the concentration of GA was 0.0053 mg/mL,the relative activity fell to 43.3 percent,its inhibition rate was 56.7%,the inhibit type was competitive inhibition,inhibition constant was 0.065 mg/mL.(5)TP can inhibit the activity of Lipase,when added 1 mg/mL of TP 1mL,the relative activity fell to 57.2 percent,its inhibition rate up to 42.8%,the inhibit type was competitive inhibition, inhibition constant was 0.17 mg/mL;TA can also inhibit the activity of Lipase,when added 1 mg/mL of TP 1mL,the relative activity fell to 47.5 percent,its inhibition rate up to 52.5%,the inhibit type was competitive inhibition,,inhibition constant was 0.107 mg/mL;GA also inhibit the activity of Lipase,when the concentration of GA was 0.0053 mg/mL,the relative activity fell to 47.5 percent,its inhibition rate up to 52.5%,the inhibit type was competitive inhibition,inhibition constant was 0.036 mg/mL.(6)This paper's results have certain differences with the same direction studies.The differences caused by that most of these studies did not eliminate the interference aider repeated tests.(7)Limited to research conditions,and study time,and other objective reasons,there is still a certain gap between the results and expected to beexplored and improved.(8)The results showed that:Drinking tea have a certain inhibit effect on the human body's intake of protein digestion and absorption,this have a certain distance between human beings expectations.But this study provide the direct scientific basis for interpretation of drinking tea especially green tea on the regulation of high-protein diet and the folk of pastoral areas prefer brick tea and some developed countries' preferences of drinking black tea with milk.(9)TP can inhibit the activity of Trypsin,The results showed that:Drinking tea have a certain inhibit effect on the human body's intake of fat digestion and absorption,this have a certain distance between human beings expectations.But this study provide the direct scientific basis for interpretation of drinking tea especially green tea on the regulation of high-fat diet.
