| PurposeTo investigate the expression of adrenomedullin(AM)protein in human epithelial ovarian carcinoma and its clinical significance.To observe the effect of adrenomedullin on the human ovarian carcinoma cells(CAOV3 cells).To provide a theoretical basis forfurther study on the role of AM in the occurrence and development of epithelial ovarian carcinoma.Materials and MethodsSelected wax lumps which were cut off from the patients on operations from Jan. 2004 to Dec.2006 at the first hospital of China Medical University,including 10 cases of ovarian normal tissue,10 cases of epithelial benign ovarian tumor(5 serous cystadenoma,5 mucinous cystadenoma)and 50 cases of epithelial ovarian carcinoma (40 serous cystadenocarcinoma,10 mucinous cystadenocarcinoma).The expression of AM were examined by the immunity organization S-P method,and the relationship between the expression of AM and the patient's clinical pathologic parameters was aggregate analyzed.The CAOV3 cells were treated with AM(1-52)of different concentration(1×10-9mol/L,1×10-8mol/L,1×10-7mol/L,1×10-6mol/L)for 24 hours, with AM(1-52)of 1×10-6mol/L for 24,48 and 72 hours.The increasing rates of cells were detected with MTT assay.The tatistical analysis was carried on by applying chi-square test and t-test.ResultsThe expression of AM exists in normal ovarian tissue and benign and epithelial ovarian carcinoma,but the expression of AM in epithelial ovarian carcinoma(74.0%) was much higher than that in normal ovarian tissue(20.0%)and benign ovarian tumor (30.0%),the difference was statistically significance(P<0.05).The expression of AM was correlated with pathologic classification(P<0.05),but not with age,clinical stage and histological types(P>0.05).With concentration of AM(I-52)(1×10-9mol/L,1×10-8mol/L,1×10-7mol/L,1×10-6mol/L),the proliferation of the CAOV3 cells could be increased(P<0.05)which showed the dose-dependent relationships.And with concentration of AM(1-52)(1×10-6mol/L),the proliferation of the CAOV3 cells could be increased(P<0.05)which showed the time-dependent relationships.ConclusionThe expression of AM in epithelial ovarian carcinoma was significantly higher than that in normal ovarian tissue and benign ovarian tumor.It was in contact with pathologic classification.AM had closed concern in occurrence and development of ovarian carcinoma.It played a key role in the progression and metastasis of epithelial ovarian carcinoma.AM is a potent stimulator for the CAOV3 cells proliferation.
