The family of fibroblast growth factors (FGFs) consists of 20 kinds of production of growth factors or oncogenes. It proceeds mitosis , differentiation growth and regeneration of neural tissue, the formation plaque of Atherosclerosis. And it has important function in the start , development and metastasis of carcinoma. The basic fibroblast growth factor (bFGF) is the focus that people are searching. The bFGF binds with its high affinity receptor , which starts the signal transduction . Then, cellular tyrosine protein kinase ( TPK ) is activated, and the tyrosine of receptor is phosphalized and some proteins in nucleus are phosphorylated or dephosphorylated.FGFs can adjust cellular function by multiple intracellular pathways , such as PLC/PKC, RTK/RAS/MAPK, JAK/STAT, PI3K/PKB. PKB is a recently found serine/threonine protein kinase. PKB is a major participant in growth factor - mediated PI3K signal transduction pathway. PI3K/PKB is activated by growth factors, such as EGF PDGF VEGF and FIGF - 1. Activated PI3K generates PtdIns(3,4,5)P3 and PtclIns(3,4)P2 PtdIns(3,4,5)P3 and Ptdlns(3,4)P2 bind to the PH domain of PKB with high affinity, which recruits PKB to plasma membrane. Ser473 and Thr308 of PKB are phosphorylated by PDK1 and PDK2. Activated PKB can phosphorylate many cellular proteins, and adjust metabolism and transport of glucose, gene transcription, protein synthesis, cell apoptosis. Excesive expression of PKB and higher activity are related to generation or prognosis of some carcinoma, and in primary gastric carcinoma, prostate, ovarian, breast carcinomas PKB is higher expression, and high expression of PKB is related to bad prognosis and low survival rate.Cyclin - CDK compound is important factor of cell cycle regulation, and lies in the checkpoint of cell cycle. The activity of cyclin - CDK compound ismainly regulated by cyclin , which express in different cell cycle checkpoint. 0-varian carcinoma is the highest morality gynecology malignant tumor. However, we are deficient of system knowledge of the effect of bFGF by PDK/PKB signal transduction pathway in ovarian carcinoma generation. In this test, we add bFGF to human ovarian carcinoma cell line CAOV3, then examine the level of PKB activity and the expression of cyclin D. We try to search the signal transduction mechanism by which bFGF regulates ovarian carcinoma cell proliferationMaterials and Methods1. Cell culture: ovarian cancer cell line CAOV3 cells were cultured in 1640 supplemented with 10% FCS with 5% C02 in air at 371.2. Tested PKB activity in CAOV3 cells treated with bFGF or wotmannin pretreatment by Western blot.3. Western blot hybridization with anti - cyclin D Ab.4. Statistical analysis with SPSS11.5.Results1. The effect of bFGF on PKB activity: the effect of bFGF (75ng/ml, 15min) is significant , and the level of PKB phosphorylated is the most highest in 10min after bFGF treated. Wortmannin the special inhibitor of PDK could decrease the effect.2. The effect of bFGF on cyclin D expression; the effect of bFGF(75ng/ ml, 15min) is the summit. Wortmannin the special inhibitor of PI3K could decrease the effect, too.Conclusion1. In the ovarian carcimona cell line CAOV3 bFGF enchances the level of PKB phosphorylation and the activity of PKB.2. The effect of bFGF on cyclin D expression is realized by PDK/PKB sinal transduction.
