ObjectiveTo investigate whether the serum from pre-eclampic women can induce the expression of intercellular cell adhesion molecule-1 (ICAM-1) on human umbilical vein endothelial cells(HUVEC) in vitro,and the effects of basic Fibroblast Growth Factors (bFGF) on the expression of ICAM-1. To discuss the protection to endothelial cellular damage of bFGF,then maybe we could find new methods to treat hypertensive disorders in pregnancyMaterials and MethodsCase selectionRandom select preeclampsia women 15 cases and normal pregnant women 15 cases from January 2006 to August2006 in our hospital. There was no significant differance in pregnant women's age and gestational weeks between these two groups. All of the cases were draw-off 10ml blood before any therapy ,and the blood were centrifuged on 3000r/min,15minutes then the supernatant was collected in freezer of -80℃.GroupThis study involved four groups:control group(Normal pregnancy women serum medium),PE group(pre-eclampic women medium),PE+b group(pre-eclampic women serum medium contain bFGF10ng/ml), negative control group(20% DMEM medium)Methods The HUVEC were incubated with medium containing serum either from women with preeclampsia or normotensive pregnancy or bFGF has been added into. Then the expression of ICAM-1 on HUVEC was determined after 24 hours by enzymelinked immunosobent assay(ELISA) technique and immunocytochemistry performed on the cell monolayer.ResultsThe result of the expression of ICAM-1 in Elisa: The average level of ICAM-1 in the PE group was 0.2751±0.01831,in control group was 0.1845±0.00761. It was higher in PE group than that in control group and found have significant differance(P<0.01). The average level of ICAM-1 in the PE+b group was 0.1879±0.01058. It was higher in PE group than that in PE+b group and found have significant differance(P<0.01).There was no significant differance between PE group and PE+b group. (P>0.05)The result of the expression of ICAM-1 in Immunocytochemical stain: the expression of positive rate of ICAM-1 in the PE group was26.59±3.577 in control group was 12.23±3.041 It was higher in PE group than that in control group and found have significant differance(P<0.01). The average level of ICAM-1 in the PE+b group was 13.75±2.913. It was higher in PE group than that in PE+b group and found have significant differance(P<0.01).There was no significant differance between PE group and PE+b group. (P>0.05)On the whole the expression of ICAM-1 was higher in PE group than that in control group and found have significant differance and was higher in PE group than that in PE+b group and found have significant differance and was no significant differance between PE group and PE+b group.ConclusionSerum from preeclampic women induces expression of intercellular cell adhesion molecule-1 (ICAM-1) on cultured HUVEC,which cue there maybe was some factors inducing endothial cells activation and injury and bFGF decrease the ICAM-1 expression on HUVEC. It directions that the bFGFcould pretect the endothelial cells which have been injured by serum from preeclampic women. Then maybe we could find new method for treating hypertensive disorders in pregnancy.
