The myocardial ischemia-reperfusion injury (MIRI) is the myocardial tissue injury induced by reperfusion after ischemia. Characteristics of reperfusion injury induced myocardial ischemia involve: myocardial stunning, arrhythmias and cell death.Therefore, it is important topic to seek safe and active traditional Chinese medicine to treat myocardial ischemia-reperfusion injury.Our country has extremely abundant resources of traditional Chinese medicine and its development perspective is capacious.YXKM Capsule is the complex traditional Chinese medicine, its major component includes Ligusticium chuanxiong, Salvia miltiorrhiza and Astragalus mongholicus. It has been proved that all the drug have the protective effects on myocardia by preclude free radical and anti-lipid peroxidation.But the effects of compatibility of them on the myocardial ischemia-reperfusion injury have not been reported at present.Method:Wistar rats, weight 240-300g, were divided randomly into six groups including sham group, ischemia-reperfusion model group, DK group (374mg·kg-1) and YXKM Capsule low dosage group (37.5mg·kg-1), YXKM Capsule middle dosage group (75.0 mg·kg-1), YXKM Capsule high dosage group (150mg·kg-1).The drugs were given by oral adminstration.The rats were anesthetized and fixed on the operating table. The chest was opened between the left third and fourth rib. The heart was exposed and coronary artery left anterior descending was ligated immediately.The heart was returned into the chest, and the chest was swiftly closed. After 60 min, the ischemia myocardium was reperfused for 12 hours. After that, the blood was collected through groin aorta and centrifuged. Then the hearts of the rats were taken out for determing the other indexes.Many parameters were determined including the activity of serum creatine kinase(CK), lactate dehydrogenase(LDH) and aspartic transaminase(AST) in serum to investigat the effect of YXKM Capsule on myocardial ischemia-reperfusion injury in rats.Myocardium of left ventricle were cut into four or five slices. Some slices were observed by electron microscope. The content of malomdialdehyde (MDA) and the activity of superoxidedismutase (SOD) were measured by colorimetry and the expresion of Bax, Bcl-2, CytC, Caspase–9 and Caspase–3 were determined by immunohistochemistry technique to approach the protective effects mechanisms of YXKM on MIRI.Result:Compared with the model group, CK, LDH and AST in serum were decreased significant by YXKM Capsule; the activity of SOD in serum activity was increased significant by YXKM Capsule and the content of MDA in serum was reduced obviously by YXKM Capsule. Compared with the model group, the change induced by ischemia-reperfusion in myocardial ultrastructure were ameliorated obviously by YXKM Capsule.Compared with the model group, the expression of Bax, CytC, Caspase-9, Caspase-3 were decreased obviously by YXKM Capsule and the expression of Bcl-2 were increased obviously.Conclusion:It was demonstrated that YXKM Capsule can inhibit the release of myocardial enzyme, protect the myocardial membrane integrity and stability by OFR be cleared and lipid peroxidation be prevented; YXKM Capsule also has the effects on anti-apoptosis by change the mitochondria membrane permeability and decrease the expression of Bax, CytC, Caspase-9 and Caspase-3, increase the expression Bcl-2. As above, YXKM Capsule can decrease cardiac myocytes in ischemia-reperfusion injury of rat, which has protective effects on myocardial ischemia-reperfusion injury in rats.
