| Background:Obesity,metabolic disorder,coronary heart disease has seriously affected the quality of life of human beings,and great degree of hazard to human life.In recent years,the metabolic syndrome whice Insulin resistance caused and coronary heart disease in the relationship between the closely watched in the major conferences.Peroxisome proliferators activated receptor(PPARs),ligand-activated nuclear transcription factor,There are three subtypes,PPAR,PPARδand PPARγ。Research shows that PPARαand PPARγis closely related to glucose and lipid metabolism,and the corresponding drug has been successfully developed.PPARδas members of the nuclear receptor superfamily,less understanding,but its widely expressed in many organs or tissues,particularly in the heart,muscle,and insulin resistance and coronary heart disease involved in inflammatory response.PPARδpromoter and 5 'untranslated region(UTR 5'2)is an important regional target gene regulation by transient transfection PPARδ5'-UTR in 3T3-F442A former fat cells.A study showed that exon 4 + 294T / C gene polymorphism near this area,so its gene polymorphism can lead to target mRNA and protein expression levels change.Study on the functional gene polymorphism help to clarify the role of PPARδgene.China's Chongqing scholars' study found that the gene polymorphism is closely related to obesity, glucose and lipid metabolism disorders and left ventricular remodeling. According to Changchun scholars,the number of metabolic syndrome was significantly higher than that of domestic in Changchun,close to the level of the United States.Therefore,we studied PPARδ294T/C on Lipids,obesity and the coronary artery stenosis.Objective:To investigate the relationship between PPARδ+ 294T/C gene polymorphism and lipid profile,obesity and the coronary artery stenosis in patients with metabolic syndrome(MS).Methods:103 patients with metabolic syndrome(based on 2004 Chinese Diabetes Association recommended standards)with coronary heart disease (MS + CDH)patients,including 61 cases of male,female 42 cases,the average age(59.46±10.51 years),72 cases of metabolic syndrome(based on 2004 Chinese Diabetes Association recommended standards)without coronary heart disease(MS)patients,including 39 cases of male,female 33 cases,the average age(56.73±9.15)years.87 cases of coronary heart disease(CHD) patients,47 cases male,female 40 cases,the average age(55.59±10.63 years), all people of Han nationality in China's Changchun.Genotyping of+294T/C variant in the PPARδgene exon region was amplified with polymerase chain reaction followed by reatriction emzyme Fok I digestion separation.The frequencies of PPARδ+294T/C genotype and allele were compared among them.Subgroups were also subdivided into of obese,nonobese, dyslipidaemic,nondyslipidaemic and coronary artery disease or non coronary artery disease according to their clinical index.Results:PPARδ+ 294T / C gene polymorphism distribution of the genotype frequencies between group difference was not significant.MS + CHD group TC,LDL-C,and CHD group compared with Normal,with a clear statistical difference.MS + CHD group CC and TC-type plasma total cholesterol and LDL-C levels were significantly higher than the TT type;CHD group and the various genotypes Normal serum lipid levels the difference was not statistically significant(P>0.05).MS + CHD group and CHD patients with multivessel disease group(≥2)was significantly higher than that in Normal group(P<0.05),single-vessel disease,MS + CHD group and CHD patients integral LAD lesions were significantly higher than those Normal group(P<0.05),while MS + CHD group and CHD group there was no significant difference between patients;LCX and RCA two points than Normal vascular stenosis patients there was no significant difference.MS + CHD group and coronary artery lesions in patients with CHD group total score was significantly higher than that Normal group(P<0.05).Analysis of PPARδgenotype and the extent of coronary artery disease and the relationship between BMI and found that different genotypes in the same group in the comparison to patients with coronary artery disease CC most obvious,MS + CHD group and CC coronary artery intimal lesions points clear higher than that of CT and TT-type(P<0.05)in the different groups,a comparison between the same gene that,in patients with type CC,MS + CHD group,CCA points lesions was significantly higher than that CHD group and Normal(P<0.05),there are statistically significant.Allele in the study,we found that in MS + CHD group,CC + allele than CT in patients with the TT-CCS increased significantly,statistical significance.Conclusions:MS + CHD group and MS patients 294T PPARδ+ / C gene polymorphism with obesity and lipid disorders close,C genotype T allele carriers than for the degree of coronary artery disease weight. |
