The theory of free radical inducing the damage of cell DNA is one of the most cancerization doctrine. Recently, there has been a considerable research interest in antioxidant capacities of phytochemicals from various materials. Several epidemiological studies suggest that a high intake of antioxidants increases the antioxidant capacity of the plasma and reduces the risk of certain, heart diseases, and stroke. Fagopyrum cymosum (Trew) Meisn is a species of buckwheat. In China, there is a long tradition of its medicinal use. The chemical researches of Fagopyrum plants discover that the Polyphenol in the plants of Fagopyrum show intensive physiologic activity of anti-tumor and oxidation,declining the blood-sugar and fat,etc. Cancer,diabetes hyperlipoproteinemia have become the serious diseases damaging human health, but the Fagopyrum plants can treat these diseases well. Hepatoma is one of the most common malignant tumor.It is insensitivity to chemotherapy and radiotherapy ,only 15% patients could radical cure because of it's diversion and palindromia.In this study we determination anti-oxidative of alcohol extracting-water precipitating extract and research the effect of extract in inhibiting the growth of cell lines SMMC-7721.Methods: (1)The antioxidant activities of Fagopyrum extracts were evaluated and compared with Vc using a 2,2-diphenyl-β-picrylhydrazyl(DPPH) assay,O2-radical scavenging method, hydroxy radical scavenging method, reducing power activity test and lipid peroxidation assay .(2) The anti-tumor effects of extract were analyzed by the MTT method,cell morphology observation (H-E),and the mechanism was studied by immunocytochemistry and flow cytometry using on hepatocellular carcinorrra SMMC-7721 cells.Results:(1)The alcohol extracting-water precipitating extract showed highest antioxidant activity.(2) MTT method showed Fagopyrum extract inhibited the growth of SMMC-7721 cells at 30ug/ml- 500ug/ml concentrations in dose-dependent manners.Under microscope observation SMMC-7721 cell displayed morphology change. Immunocytochemistry and flow cytometry showed that the extract of Fagopyrum effects on survivine expression and the cell cycle.Inhibitory in vitro activity of extract on matrix metalloproteinase(MMP) showed that the extract inhibited the MMP-16 and MMP-13 activity and the IC50 was 26.8ug/ml,9.7ug/ml.Conclusion: The alcohol extracting-water precipitating extract of Fagopyrum cymosum possess antioxidant activity and antitumor activity. Along with scientific and technological progress and the improvement of people's standard of living, antioxidants have become food, medicine and industry, such as an indispensable product. However, since the synthetic antioxidants BHA, BHT and other side effects after being confirmed for natural antioxidants work in recent years, scientists have become a major topic of research. Thus find natural, and efficient, low toxicity of antioxidants become an inevitable trend. Chinese herbal medicine is an important natural source of antioxidants. So isolate from traditional Chinese medicine, extracting more antioxidants are broad-based and application prospects. Furthermore, antioxidants prevent cancer significant role for the prevention and treatment of oncological diseases opened a new idea. In the future anti-cancer drugs, non-toxic, no side effects of anti-oxidants and effective and lasting way to supply should have greater value. The choice of buckwheat as the research object, using five different methods on the antioxidant activity of compounds comprehensive evaluation, in addition, the choice of human hepatoma cell line SMMC-7721, the water-alcohol extract of buckwheat by the law compounds in vitro and tumor suppressor activity of inspection by flow cytometry, immunohistochemistry, activity of matrix-examine its anticancer mechanism. This study mainly provid a certain theoretical foundation and experimental evidence for seeking natural antioxidants and anti-cancer drugs.
