| Objective :To study the expression of HIF-1αand VEGF in the tissue of normal cervical tissues,CIN and cervical cancer tissues and metastatic lymph node.To explore the correlation between the expression of HIF-1αand VEGF and the age of patient, the tumour's magnitude, the cervical cancer status, including pathological types,clinical stage, infiltration, Tumor necrosis and differentiation, and prognosis. The expression and the clinical significance of histoplasma tissue inhibitory factor in cervix epithelial tumor tissues.Methods: To select randomly 108 cases of cervical specimen who were operated at the Department of Obstetrics and Gynecology at the China-Japan Union Hospital of Jilin University and the third Hospital of Jilin University(the second portion) in 2006-2008. All the specimens of cervical cancer were selected from the tissues without adjuvant radiotherapy and chemotherapy,including 16 cases of normal cervical tissues,38 cases of CIN and 54 cases of cervical epithelial cancer tissues and 15 cases with metastatic lymph node. which were divided into several groups according to pathological types, clinical stage and differentiation. metastatic lymph node or metastatic omentum. The expression of HIF-1αand VEGF were detected by immunohistochemistry SP method. Results of the reactions and analysis of the clinical course of the patients were subjected to statistical analysis.Result:â‘ T he positive rates of HIF-1αand VEGF were 0%,0% in normal cervical tissues,5.8%,11.1% in the CINâ… -â…¡tissues, 40%,50% in the CINâ…¢t issues, 66.6%,72.2% in cervical cancer, respectively. The CINâ…¢tissues group differed significantly from normal cervical tissues and the CINâ… -â…¡t issues by the expression of HIF-1αand VEGF. And both groups significantly differed from the cervical cancer group. A significant positive correlation was found between the expression of HIF-1αand VEGF in the cervical cancer group. The expression of HIF-1αand VEGF were higher in the cervical cancer than in the CINâ…¢tumor. There was significant difference between two groups statistically(p<0.05).The brown granules of VEGF positive cell were mainly distributed in cytoplasm, the brown granules of HIF-1αpositive cell were mainly distributed in nuclei and cytopasm.â‘¡The positive rates of HIF-1αand VEGF in cervical cancer tumour's magnitude diameter≤2cm were 38.9%,50%,that in diameter 2-5 cm were 75%,79%, that in diameter≥5 cm were 91.6%,92.0%respectively. The expression of HIF-1αand VEGF in bigly tumour's magnitude diameter was higher than that in smallly (P<0. 05).â‘¢The positive rates of HIF-1αand VEGF in cervical cancer clinical stageâ… a were 53.1%,59.4 %,that in clinical stageâ… b were 83.3%,88.9%,that in clinical stageâ…¡w ere 100%,100% respectively. The expression of HIF-1αand VEGF in advanced stage(â…¡) was higher than that in early stage(Ib,Ia)(P<0. 05).â‘£The positive rates of HIF-1αand VEGF were 93.3%,93.3% in cervical cancer tissues with cases of metastatic lymph node 51.4%,64.1% in cervical cancer without cases of metastatic lymph node or metastatic omentum. There was significant difference between two groups statistically(p<0.05).⑤The positive rates of HIF-1αand VEGF in cervical cancer Tnmor non-necrosis were 50.0%,57.1 %,that in Tnmor necrosis were84.6%,88.5%respectively. The positive rates of HIF-1αand VEGF were significantly higher in Tnmor necrosis than in cervical cancer Tnmor non-necrosis. There was significant difference between two groups statistically(p<0.05).â‘¥the positive expression of HIF-1αand VEGF in squamous carcinoma of the cervix were 71.4%,76.2%,that in adenocarcinoma of the uterine cervix were 50.0%,58.3%, there was no significant difference between two groups statistically(p>0.05).⑦The positive rates of HIF-1αand VEGF in cervical cancer well differentiation and moderately differentiation were 60.0 %,70.0 % , that in moderately differentiation were 66.6%,70.8%,that in poorly differentiation were70.0%,75.0 % respectively.The positive rates of HIF-1αand VEGF were significantly higher in poorly differentiation than in well differentiation and moderately differentiation. But there was not significant difference between two groups statistically(p>0.05).â‘§T he positive expression of HIF-1αand VEGF in the age of patient<40 were 59.4% 65.6% ,in the age of patient≥4 0 were 77.3%,81.0%.There was not significant difference between two groups statistically(P>0.05).⑨The positive expression of HIF-1αand VEGF in cervical cancer infiltrating superficial muscular layer were 68.4% 78.9% ,in infiltrating profunda muscular were 65.7%,68.6%.There was not significant difference between two groups statistically(P>0.05).â‘©There was a positive correlation between the positive rates of VEGF and the positive rates of HIF-1α. Conclusions:1. HIF-1αand VEGF were not expressed in normal cervical tissue, were low expressed in CIN tissue and were high expressed in cervical cancer.It hints that HIF-1αand VEGF play an significant role in carcinogenesis and growth of cervical cancer.2. The more later in clinical stage and the more biger in the tumour'magnitude, the more higher positive rates of HIF-1αand VEGF were. The positive rates of HIF-1αand VEGF were correlated with tumour'magnitude and clinical stages of cervical cancer ,but were uncorrelated with in celldifferentiation,infiltrating superficial muscular layer ,the infiltration and the pathological type.3. HIF-1αand VEGF were positively correlated with the development of cervical cancer.4. The positive rates of HIF-1αand VEGF was higher in cervical cancer tissues with metastatic lymph node and Tnmor necrosis more than without metastatic lymph node and Tnmor necrosis.The expression of HIF-1αand VEGF was correlated with metabasis.5. We can judge malignant degree of the tumor on monitoring the expression of HIF-1αand VEGF, We still need to further study vasoformative regulation mechanism of cervical cancer in molecular level, it will provide a new way of inhibition and interception vasoformative and future gene therapy for cervical cancer offer theory . |
