The supersonic assistant extraction optimization technics parameters of Se-enriched green tea ethanolic extracts were studied. The antioxidant activity of Se-enriched green tea ethanolic extracts was evaluated. The antitumor activity (in vitro) on A549, HepG2 and HeLa cell lines and the composition of Se-enriched green tea ethanolic extracts were researched. The results were shown as follows:The effects on yield of ethanol concentration, ratio of solid/liquid, temperature, time, power of supersonic and times were researched by orthogonal design of L9(34). The antioxidant activities of different treatments were determined. The product of treatment 2 showed excellent antioxidant activity and yield. The technics parameters of treatment 2 were confirmed as the optimization technics parameters of Se-enriched green tea ethanolic extracts: Se-enriched green tea was extracted at 50℃in 50% ethanol for 1.5 hour two times, with 1:15 as ratio of solid/liquid and 250 W as power of supersonic.The Se-enriched green tea ethanolic extracts showed significantly higher antioxidant activity than Se-enriched green tea aqueous extracts andα-tocopherol at the same concentration.Se-enriched green tea ethanolic extracts possessed significantly inhibition effect on A549, HepG2 and HeLa cell lines by MTT method. The inhibition effect showed dosage relation. The inhibition effect on A549 cell lines was significantly stronger than than that of regular green tea ethanolic extract and was not significant different from positive control 5-FU. The inhibition effect on HepG2 cell lines was significantly stronger than than that of regular tea ethanolic extract, regular green tea ethanolic extract + Na2SeO3 and positive control 5-FU. The inhibition effect on HeLa cell lines was significantly stronger than than that of Se-enriched green tea aqueous extracts and positive control 5-FU.Se-enriched green tea ethanolic extracts mainly contained selenium compounds 4.35±0.11μg·g-1, polyphenols 558.44±2.31 mg·g-1, protein 257.52±4.05 mg·g-1, polysaccharide 133.73±3.02 mg·g-1, and theine 59.94±0.01 mg·g-1. Selenium and polyphenols may mainly contribute to the anticarcinogenic and antioxidant activities of Se-enriched green tea ethanolic extracts. The content of selenium and polyphenols of Se-enriched green tea ethanolic extracts were significantly higher than Se-enriched green tea aqueous extracts. It could conclude that extraction of functional component from Se-enriched green tea with supersonic with 50% ethanol as solvent was more efficient than extraction with water.
