Objective To study the significance of the change of T cell subsets in different MHV-3 induced viral hepatitis.Methods 100pfu of MHV-3were injected intraperitoneally to 16 mice (8 Balb/cJ ,8 A/J) ,and then the T cell subsets ratios of total T lymphocytes in blood,spleen and liver were examined at 0,24,48 and 72 hours post MHV-3 infection by flow cytometry. DN T cell and CD8~+ T cell were isolated form spleen cell suspension by immunomagnetic beads and the cytotoxicity of DN T cell were tested by MTT assay.Results CD3~+CD4-CD8- T cell ratio in Balb/cJ mice raised significantly since 24 hours post MHV-3 infection and reached the peak until day3.CD3~+CD4~+CD8- and CD3~+CD4-CD8~+ T cell ratios degraded accordingly. However,the T cell subsets ratios in A/J mice have no significant changes. Furthermore, a dose-dependent inhibitory effect of CD3~+ DN T cells on the proliferation of syngeneic CD8~+ T cells was evidenced.Conclusions The primary different DN T cell ratios ,their prognosis post MHV-3 infection in between Balb/cJ and A/J mice and the inhibitory effects on synteneic CD8~+ T cells suggested that DN T cell plays an important role in the process of the development of fulminant viral hepatitis.
