ObjectiveAquaporins(AQPs)expressed in various organs and tissues of the body, AQP1,which was mostly investigated played an important role in the physiological and pathological processes.Inflammation,stress,lung injury or ARDS could induce the changes of AQP1 expression,so did many pathogenic bacteria toxin and virus;It had been shown that AQP1 was involved in the proliferation and apoptosis of many cells, but it was still controversial.In our experiment we determined the role of Mycobacterium tuberculosis polypeptide antigen(MTb-Ag)on the Balb / c 3T3 fibroblasts by detecting the changes in morphology and the expression of AQP1.MethodsCells was divided into control group,MTb-Ag 10μg/ml group and MTb-Ag 30μg/ml group,the expression of AQP1was detected by Western blot and Immunofluorescence.ResultsCompared with the normal cell group,the cell growth rate was accelerated markedly in MTb-Ag 10μg/ml group from one to three days,while cells in MTb-Ag 30μg/ml group almost dead till the third day.AQP1 expression probed by immunoblot increased significantly in MTb-Ag 10μg/ml group over time and immunofluorescence assay also got same results in the group.Interestingly,there was not AQP1 induction in MTb-Ag 30μg/ml group. DiscussionMTb-Ag 10μg/ml stimulate the growth of Balb/c3T3 fibroblasts.Fibroblasts ultured in 30μg/ml MTb-Ag were almost dead on day 3.We speculate that 30μg / ml of MTb-Ag may be toxic to cells;Western Blot and immunofluorescence confirmed the results that AQP1 expression was significantly increased in MTb-Ag 10μg/ml.We supposed AQP1 was likely involved in the regulation of Balb/c 3T3 cell proliferation.Further studies were needed to confirm it.ConclusionMycobacterium tuberculosis components increased the expression of AQP1 and stimulate Balb/c3T3 fibroblast proliferation,providing a potential link between AQP1 and the early formation of tubercler.
