Objective To establish an animal model of corneal blood staining in rabbit, and observe the pathologic features of it.Methods 12 rabbits were destructed the endothelial cells and injected blood in the anterior chamber repeatedly, the corneas were observed daily. Designed the 24 eyes for three stages(â… ,â…¡,â…¢) corresponding the 3 of development of cornea blood staining that the staining occupied 1/3,2/3 and almost the whole stroma of cornea. Cut off the corneas when they had significant clinical appearances of each of the three stages for pathological examinations-HE,Perl's Prussian blue staining and transmission electron microscopic(TEM) examination.Results In average 2.2,3.5 and 7.4 d formedâ… ,â…¡,â…¢stage of corneal blood staining respectively. Both HE staining and Perl's Prussian blue staining reflected that blue Particles increased gradually amongâ… -â…¢stage. Perl's Prussian blue staining disclosed intracellular hemoglobin particles inâ…¡stage and extracellular as well as intracellular hemoglobin particles inâ…¢stage in keratocyte. TEM examination showed the necorsis of keratocytes in bothâ…¡andâ…¢stages, and revealed the lysis of collagen fibers and the formation of fibroblast.Conclusion The rabbit model may be performed easily and repeatedly with less complications, and was similar to the clinical process of corneal blood staining after traumatic hyphema. It can be useful for the study of pathogenesis and medical intervention of corneal blood staining. Objective To study the therapeutic effects of 0.37%EDTA -1.0%chitosan eye gel on corneal blood staining.Methods Corneal blood staining model ofâ…¢stage of 12 New Zealand white rabbits were made. The 24 blood stained corneas and were divided into 3 groups and then divide equally each cornea for two parts: the one part of corneas were put in saline,0.37%EDTA eye drop and 0.37%EDTA -1.0% chitosan eye gel 3ml for 35 days( take out 0.3ml every 5 days for the measure of concentration of pricipitated iron-ion);the other part of corneas and the corneas soaked for 35 days were proceed for pathological examinations.Results 0.37%EDTA -1.0% chitosan eye gel group chelated markedly higher iron-ion concentration than the other two groups(P<0.001),the soaked liquid had less debris, the corneas had more integrated epithelium,more compact collagen layers and less hemosiderin.Conclusion 0.37%EDTA -1.0% chitosan eye gel has strong effect on chelating iron-ion of corneal blood staining, and may be effect drug on the therapy of corneal blood staining.
