Objective To investigate the influence of Hhcy on vascular endothelial function and inflammation.To explore the effect mechanism of atorvastatin and fosinopril on endothelial function which was damaged by hyperhomocysteinemia.Methods Fifty male SD rats were divided randomly into five groups,each group involved 10 rats.Control group were fed with standard rat diet and were poured water into stomach every day; methionine group were fed with 3% methionine diet and were poured water into stomach every day; atorvastatin group were fed with 3% methionine diet and were poured atorvastatin 10mg.kg-1 into stomach every day;fosinopril group were fed with 3% methionine and were poured fosinopril 10mg.kg-1 into stomach every day;union group were fed with 3% methionine diet and were poured atorvastatin and fosinopril 10mg.kg-1 into stomach every day. After 8 weeks,the levels of IL-8 and nitric oxide in serum were determined. In the same time, the levels of Hcy,SOD,and MDA in plasma were analyzed.Results 1.The levels of homocysteine:after 8 weeks, the levels of homocysteine in control group were lower than other groups(P<0.001) which were not different from each other(P>0.05).2.The levels of IL-8: the levels of IL-8 in control group were lower than other groups, methionine group were higher than other groups(P<0.001), atorvastatin group were lower than fosinopril Group (P<0.01) and were not different from union group(P>0.05), fosinopril group were higher than union group( P<0.001 ) .3.The levels of NO: control group were higher than other groups(P<0.001), methionine group were lower than atorvastatin group, fosinopril group and union group(P<0.001); atorvastatin group were higher than methionine group and fosinopril group(P<0.01), atorvastatin group were not different from union group(P>0.05),fosinopril group were higher than methionine group(P<0.001) and lower than other groups(P<0.001).4.The level of SOD: control group were higher than other groups(p<0.001), atorvastatin group were not different from fosinopril group and union group. Fosinopril group were lower than union group.5.The level of MDA:control group were lower than other groups(P<0.001), methionine group were higher than other groups(P<0.001), atorvastatin group were lower than fosinopril group(P<0.05) and higher than union group (P<0.05), fosinopril group were higher than union group(P<0.001).Conclusions1. Hhcy can be induced after methionine-rich diet in rats.2. Hhcy can induce endothelial dysfuncion by inhibiting the production of NO and activation of inflammatory reaction and oxdize stress.3. Atorvastatin and fosinopril can not chang the levels of homocysteine in plasma. However,they can protect endothelial function destroyed by Hhcy.If they were used at the same time,the function would be enhanced.
