The Preventive Of Atorvastatin On Atherogenesis Induced By Hyperhomecysteinemia In Rats

AIMS:Hyperhomecysteinemia(Hhcy) is an independent risk factor of atherosclerosis(AS). Statins in cardiovascular aspects of pleiotropic caused people to pay attention in recent years. In this study, we set up a rat Hhcy model induced by homomethionin diet.With the influence of high and low dose atorvastatin,measuring the level oft Hcy,ox-LDL,MDA in rat bloods and the express of NF-κB P65,LOX-1 mRNA,observing pathologica change of aorta by HE staining, analysising difference between groups,to explore atorvastatin to Hhcy rats myocardial protection.METHODS:The rats were divided into four groups randomly,each of which was 8.The four groups include:①normal control group(control group):fed by elementary animal feeds+0.9%Nacl(5ml/kg/d);②methionine group(Hcy group):fed by elementary animal feeds,adding 1.7% methionine+0.9%Nacl (5ml/kg/d);③methionine+low dose Atorvastatin(low-dose group):fed by elementary animal feeds,adding 1.7% methionine+ Atorvastatin(2.5mg/kg/d)④methionine+high dose Atorvastatin(high-dose group):fed by elementary animal feeds,adding 1.7% methionine+ Atorvastatin(10mg/kg/d). we set up a rat Hhcy model induced by homomethionin diet after 8 weeks. The level of tHcy in plasma were measured by using the method of circulation enzymatic;To measure ox-LDL by ELISA method;to measure MDA by thiobarbituric acid reactive substance method;to measure the expression of NF-κB P65 in Myocardium by Westorn blot method;to measure the expression of LOX-1 by RT-PCR;to observe pathologica change of aorta by HE staining.RESULTS:①The levels of tHcy in blood plamsa on control group were obviousbly lower than other groups,which were not different from each other.(p>0.05)②The level of ox-LDL and MDA on atorvastatin groups were lower than Hcy group,and high dose atorvastatin group were lower than low dose group.③The expression of NF-κB P65 and LOX-1 in myocardium on atorvastatin groups were lower than Hey group,and high dose atorvastatin group were lower than low dose group.④Hcy group caused the pathological morphology change,but atorvastatin group reduced the damage.CONCLUSION:Hhcy by high methionine diet could cause AS. Atorvastatin can reduced the Hhcy-induce atherosclerosis damage,and with doses of increase,it could be more protect the Myocardium Against Hyperhomecysteinemia.
