Experimental Study Of Radiated Xenogenic Acellular Dermal Matrix Incorporated With Drug

Objective The preparation of xenogenic acellular dermal matrix(xeno-ADM) incor- porated with drug. To observe the graft immunological rejection of the xeno-ADM,To research histocompatibility of tissue engineered skin and explore a good idea in the treatment of full-thickness skin in juries or serious wound .Method Using 1mol/L Hypertonic saline and 0.25﹪Trypsin and 0.5% TritonX-100 to prepare xeno-ADM, After that the xeno-ADM was treated with 1200mg/L triclosan and 60CO radiation to prepare xeno-ADM incorporated with drug. The xeno-ADM incorporated with drug was examined by microbial examination and exmined macroscopically by optical electronic microscope and scanning electron microscopic technique . 30 abult rabbits were randomly divided into three groups according to different skin grafts,i.e. split-thinkness skin autograft only(group A,n=10),xeno-ADM compsited with split-thinkness skin autograft(groupB,n=10),xeno-ADM incorporated with drug composited with split-thinkness skin autograft (groutC,n=10). Three groups were examined by lymphocyte proliferation activity and examined content of cytokine IL-10,IL-2.The survival rate, the contraction degree of the grafts and the histogical changes in grafting were observed at 2,4,6,8,12weeks after the operation.Results observed under light microscopy, the epidermis was completely removed, there were not any cellular components,hair,follies, sebaceous glands and vessels in the dermis to be foud.The bacteriostasis test of groupC is obviously higher than other groups(P<0.05);GroupA:OD0.1783±0.0325,groupB:OD0.1801±0.0540,groupC:OD0.1732±0.0326.The result of the lymphocyte proliferation activity exhibited no differece in three groups(P>0.05).The expressions of IL-2 and IL-10 were no obvious differece in three group(P>0.05);After the operation ,the contraction degree of A group grafts were severity, there were full differentioation of epithelium orderly collagen proliteration and slight reconstruction of the epithelial-dermal conjunction structure and the remodeling the basement membrane in groups B,C.Conclusion The xenogenic acellular dermal incorporated with drug is an ideal dermal substitute ,which removed completely the epidermis and cellular components in dermis and retained the integrity structure of collagen fibers,and which have partial,good and strong bacteriostatic effect.The tissue engineered skin has favorable histocompatibility and doesn't arose obvious immunological rejections.
