Objective To determine the protective effect of magnesium fortified drinking water on experimential atheroselerosis formation in rats.Methods Thirty male Wistar rats were divided randomly into three groups,each group involved 10 rats.Model group were fed with high-fat diet and normal water; high-magnesium group with high-fat diet and magnesium fortified water of high density (50mg/ml),low-magnesium group with high-fat diet and magnesium fortified water of low density(25mg/ml). At the end of 8 weeks,the levels of blood lipids were determined by immunoradiometry methods ,the level of IL-8(Interleukin-8) in serum was determined by ELISA methods,and immunohistochemical methods were used to detect the expression of NF-κB(nuclear transcription fator-κB) in endothelial cell and smooth muscle cell of aorta.Results 1.The levels of blood lipids.The levels of TC and TG in high-magnesium group and low-magnesium group were lower than model group(P<0.001) . The level of LDL-C in high-magnesium group and low-magnesium group were lower than model group(P<0.01) And there was no different of the level of HDL-C among three groups (P>0.05). 2.The positive rates of NF-κBp65.About the positive rate of NF-κBp65 in endothelial cells and smooth muscle cells of aorta,the rate in model group was higher than that in high-magnesium group (P<0.01),and higher than that in low-magnesium group (P<0.05).The rate in high-magnesium group was lower than that in low-magnesium group (P<0.05). 3.The levels of IL-8.the level of IL-8 in model group was higher than the other groups (P<0.001),low-magnesium group was higher than high-magnesium group(P<0.01).Conclusions1. Early atheroselerosis formation can be induced by high-fat diet and vitaminD3 injecting in rats.2. Fortified magnesium can decrease the levels of blood lipids and release atheroselerosis formation.3. Magnesium can depress the activation of of NF-κBp65 and expression of IL-8 in endothelial cells and smooth muscle cells of aorta,which maybe correlate with releasing atheroselerosis formation.
