Objective:The purpose of this study was to investigate the effects of magnesium copper(Mg-Cu)alloy,as cast pure magnesium(Mg)and titanium(Ti)alloy on the growth and apoptosis of H22 tumor tissue in mice,observe the degradation of metal implanted in vivo,and explore the effects on the expression of tumor acid-base related protein carbonic anhydrase IX(CAIX)and hypoxia inducible factor-1 α(HIF-1 α).To evaluate whether degradable magnesium metal and its alloy have inhibitory effect on H22 tumor in mice,and to explore the possible mechanism.Methods:In this experiment,H22 Tumor bearing mice model was used,and the experiment was started when the longest diameter of tumor reached 0.8 cm.The tumor weight and volume were monitored on the 3rd,6th,9th,12 th and 15 th day after implantation of Mg-Cu alloy,pure Mg and Ti alloy.On days 3,6,9,12 and 15,the mice were anesthetized and prone on the X-ray table of soft tissue of molybdenum target.The degradation of metal in vivo was observed by molybdenum target photography,and the surface changes of metal extracted from tumor tissue were observed by scanning electron microscope.On the 15 th day after the operation,the mice were killed and the tumor tissues were dissected.H & E staining,TUNEL staining and immunohistochemical staining were used to detect the apoptosis and expression of related proteins.All data was expressed as mean ± standard deviation(` x±SD).SPSS17.0 software and Graph Pad Prism 5.0software were used for statistical analysis.Analysis of variance and Bonferroni pairwise mean comparison were used for statistical analysis.P < 0.05 showed that the difference was statistically significant.Results:Compared with Ti alloy group,the growth of tumor in Mg-Cu alloy group and pure Mg group was inhibited.The average volume and weight of tumor in Mg-Cu alloy and pure Mg group were significantly lower than those in titanium alloy group at 9,12 and15 days after operation(P < 0.05).The results show that the degradable metal Mg and MgCu can degrade and produce gas in mouse H22 tumor over time.The Mg-Cu alloy has been completely degraded in three days.The surface degradation of pure Mg is more and more serious with time,and the cracks are more and more dense.On the 15 th day,the apoptosis rate of tumor tissue in the mice implanted with Mg-Cu alloy and pure Mg was significantly higher than that in the mice implanted with Ti alloy(P < 0.05);compared with the mice implanted with Ti alloy,the expression of CAIX protein in the tumor tissue implanted with Mg-Cu alloy and pure Mg was significantly down regulated(P < 0.05),and the expression of HIF-1 α was changed,but there was no statistical significance(P > 0.05).Conclusion:Degradable magnesium and its alloy can inhibit the growth of H22 tumor in mice and affect the expression of acid-base related proteins.
