Bladder cancer is the most common cancer of genitourinary tract in china. The majority is the superficial bladder cancer. The treatment by surgery is the transurethral resection or laser vaporization, or partial cystectomy. After the surgery, intravesical chemotherapy is often undertaken. The most common agents are bacillus Calmette- Guerin vaccine, mitomycin C,hydroxycamptothecin,etc.Brucea javanica oil emulsion (BJOE) is a new agent of the traditional Chinese medicine developed by Shenyang Pharmacy College. It has been put into clinical practice from the 1970s and showed satisfactory anti-cancer effect. Irrigation of bladder with Brucea javanica oil emulsion has some advantages, such as low recurrence rate, few side effects and well tolerance,etc. So it has great significance to investigate the mechanism of BJOE for the therapy of bladder cancer. This study observed the effect of different concentration BJOE on proliferation of J82 cell and the possible mechanism involved.The inhibitory effect of BJOE on J82 cell proliferation was measured by MTT assay, Hoechst 33342 treated J82 cells were observed by fluorescence microscope. Cell apoptosis was detected by morphological observation and flow cytometer analysis, and Caspase-3 activity was assessed using a Caspase-3 kit.0.0~4.0 ml/L Brucea javanica oil emulsion inhibited J82 cell proliferation in a dose-dependant manner.2.0 ml/L Brucea javanica oil emulsion induced typical apoptosis after 24 h of incubation. The Caspase-3 activity of 2.0 ml/L Brucea javanica oil emulsion group was higher than those of control group or Brucea javanica oil emulsion plus Ac-DEVD-CHO group.The results indicate that BJOE can inhibit the proliferation of J82 cell in vitro,it can induce apoptosis. Caspase-3 activation was detected which indicatesd that BJOE may induce J82 cell apoptosis through Caspase-3 pathway.
