Objective:To ascertain the pharmacological effect of traditional Chinese medical compound naozhongxiao that can intervene the cerebral edema in rats with focal cerebral ischemia. And to probe and analyse the action mechanism of naozhongxiao in many ways in cerebral edema at the acute ischemic apoplexy stage. The results of the study provided an experimental basis for developing the clinical application of naozhongxiao.Methods and Contents:It had been erected that the model of focal cerebral ischemia with suture-occluded method in rats in order to observe the effects of naozhongxiao on the content of cerebral water, Ca2+, phospholipase A2(PLA2). Results:1. The PLA2 activity, cytosolic free Ca2+ concentration in neuronal cells,and brain water content were progressively increased from 30 minutesto 120 minutes after middle cerebral artery occlussion.2. The brain water content was positively correlated with the cerebralcortex PLA2 activity and neuronal cells cytosolic free Ca2+ concentration in 120 minutes after ddle cerebral artery occlussion.The cerebral cortex PLA2 activity was positively correlated withcytosolic free Ca2+ concentration in neuronal cells.3.Naozhongxiao and Nimodipine could decrease to some degree the content of cerebral water, Ca2+ ,PLA(P<0.05) and increase the conten of Ca2+. Compared with model group 4. Nimodipinesimilar to Naozhongxiao can significantly decrease PLA2 activity, cytosolicfree Ca2+ concentration in neuronal cells, brain water content after cerebralischemia and alleviate brain edema. Conclusion:1. PLA2 activation is involved in the process of cerebral cell ischemi injury .The severity of neuronal cell injury is closely related with thePLA2 activity.2.Increased cytosolic free Ca2+ concentration in neuronal cells after cerebral ischemia activates PLA2 and deteriorates cell injury.3.Naozhongxiao showed markedly preventive effect on cerebral edema in focal cerebral ischemia rats. The mechanism could be concerned with relieving the energy dysmetabolism, acommodating the ionic balance of exterior and interior of cell and decreasing the PLA2 level in brain. The other pharmacological mechanisms have yet to be investigated. Innovations: 1. The guiding ideology of naozhongxiao is constituted of six methods, which will give full play the curative effects to the series ofchanges in cerebral edema in each various phase at the acute ischemic apoplexy stage. 2. To aim the curative effects in cerebral edema at the acute ischemic apoplexy stage. The study is how to apply the traditionalchinese medicine in emergency treatment field.
