Intervention Effect Of Naozhongxiao In Treating Cerebral Edema With Acute Focal Cerebral Ischemia In Rats

Objective: To ascertain initially if Naozhongxiao could intervene the cerebral edema with acute focal cerebral ischemia in rats. Methods: Wistar rats were randomly allocated into five groups. Naozhongxiao 15g/kg group and 30g/kg group were orally administered herb liqids at 1.5ml/kg capacity and different concentrations ingestlibly. By the same way, Sham operation group and model control group were taken with normal saline.Positive medicine control group was taken with conversed Weinaolutong liquid. The process continued for seven days, one time every day. One hour after ingestion in the seventh day, middle cerebral artery(MCA) occlusion was performed with suture-occluded method to cause brain edema. Four hours after MCA occlusion, the alive rat behavior was scored and scaled. The rats that were effective models were taken blood and brains removed for the determination of cerebral coefficient and water content, morphology, blood viscosity and red cell aggregation index, cerebral MDA content and SOD activity. Results: The cerebral coefficient and water content, blood viscosity and red cell aggregation index, cerebral MDA content of the model control group were significantly higher than those in sham operation group (p<0.01 or p<0.05). The cerebral SOD activity of the model control group were significantly lower than that in sham operation group (p<0.01). Both Naozhongxiao 15g/kg group and 30g/kg group, Positive medicinecontrol group could significantly attenuated cerebral coefficient and water content, blood viscosity and red cell aggregation index, cerebral MDA content(P<0.05), and ameliorate SOD activity(P<0.05)when compared to the values of model control group. Except the sham operation group,the other group all cerebral tissue were observed edema change by optical microscope. Conclusion: Naozhongxiao can prevent the progress of cerebral ischemic edema following MCA occlusion in rats, and to a certain extent directly give effect to alleviate turbidity and noxiousness, reduce phlegm and dampness, eradicate extravasated blood, and assist the healthy and weak.
