Empirical Study On The Mechanisms Of Anti-angiogenesis And Anti-inflammation Of Thalidomide And Traditional Chinese Herbs On Rats With Collagen Type â…¡ Induced Arthritis

Objectives:To investigate the effect of thalidomide and traditional Chinese herbs on rats with collagen type II induced arthritis (CIA); To study the mechanisms of anti-angiogenesis and anti-inflammation on rats with CIA.Subject:Rat SD.Methods:At first, the model of rats with CIA was induced by injecting intradermal collagen II and complete Freund's adjuvant (CFA). Next day, the therapeutic administration of thalidomide (Thd, 150 mg·kg-1·day-1×6 weeks) and traditional Chinese herbs (40 g·kg-1·day-1×6 weeks) was given to the rats of treatment group, respectively. The rats of control group and model group were administrated with phosphate buffered saline in the same dose.The mean arthritis index (MAI) was scored everyday. The rats were sacrificed at the sixth week and knee joint from rats were estimated in morphology and immunohistochemistry assay in order to detect microvessel density (MVD) of synovial membrane. Then, the sera was drawn, also the concentration of interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α), vascular endothelial growth factor (VEGF), and matrix metalloproteinases-1,2,3,9 (MMP-1,2,3,9)in sera were detected by Western blotting. Results:1. The model of rats with CIA induced with intradermal collagen II and complete Freund's adjuvant, which detected by the mean arthritis index (MAI) and immunohistochemistry, was verified to be successful.2. The arthrocele of rats with CIA was obviously; The mean arthritis index (MAI) and the expression of IL-1βand TNF-αin model group were higher than the other group; Thalidomide and traditional Chinese herbs can significantly decreased the expression of IL-1βand TNF-αin sera.3. Synovial tissue of joint and the number of new blood vessels in rats with CIA increased significantly. Thalidomide and traditional Chinese herbs can restrain the hyperblastosis of synovium to decrease the mean arthritis index by restraining the hyperblastosis of synovium and the number of new blood vessels in rats with CIA. To investigate deeply, the administration of thalidomide and traditional Chinese herbs significantly decreased the expression of VEGF and MMP-1,2,3,9 in sera.Conclusions:1,The model of rats with CIA induced with intradermal collagen II and complete Freund's adjuvant was verified to be successful.2. The expression of IL-1βand TNF-αintimately correlates with Inflammatory reaction; They may participate in the pathogenesis of inflammation of RA.Thalidomide and traditional Chinese herbs can obviously improved the mean arthritis index of rats with CIA by reducing the expression of IL-1βand TNF-α.3. Thalidomide and traditional Chinese herbs had an inhibitory effect on angiogenesis of rats with CIA by restraining the expression of VEGF and MMP-1,2,3,9.
