introduction The Radiation-induced damagement to heart usually include acute/chronic pericarditis, accelerated atherosclerosis(such as coronary artery disease), conduction abnormalities, valvular changes, and, notably, pericardial and myocardial fibrosis. On the other hand, Angiotensin II (AngII) and aldosterone are the key constituents of the renin-angiotensin-aldosterone-system and play an important pathophysiological role in cardiovascular disease. Synthesis of aldosterone and angiotensin II will increase when there is trouble in hearts. The pathophysiology of RIHD is similar to that induced by angiotensin-aldosterone.we irradiated Sprague-Dawley rats with a single dose of 0,15 or 18 Gy locally to the heart. 3 months after irradiation, the concentrations of angiotensin II and aldosterone were determined by using ELISA means. The relationship between angiotensinII -aldosterone and RIHD will be discussed in this study, and the pathophysiological effect of angiotensin II -aldosterone in the progress of radiation-induced heart disease will also be analysed.materials and methods1 .tissue samplesSprague-Dawley rats (3 months old, 190-210g, male = female,buying from the animal centre of China Medical University) were excuted by over-anaesthetization with 3.6% chloral hydrate . 3 months after irradiation, the heart tissue was excised and stored in refrigerator at the temperature of -85℃.2.reagentsAngiotensin II and aldosterone ELISA assay kits were purchased from RapidBio Lab.3. methodsSprague-Dawley rats were irradiated with a single dose of 0, 15 or 18 Gy locally to the heart. 3 months after irradiation, the concentrations of angiotensin II and aldosterone were determined by using ELISA. SPSS10.0 statistical software was applied for data analysis. Results are expressed as means±SE. Data were analyzed by t-test. Differences were considered statistically significant at a value of P < 0.05. resultsCompared with non-irradiation group, the angiotensin II concentration in irradiation 15Gy group and18Gy group have significant difference. P<0.05. The concentration of aldosterone in irradiation 15Gy group and18Gy group have significant difference. P<0.05.ConclusionsThere have relationship between angiotensinII -aldosterone and radiation-induced heart disease. .Angiotensin II -aldosterone plays an important pathophysiological role in the progress of radiation-induced heart disease. What's more, with the radiation doses rise, the impacts of angiotensin II -aldosterone on radiation-induced heart disease may be stronger.What extent of the relationship between angiotensin II-aldosterone and radiation-induced heart disease needs to deeply study.
