IntroductionCodeine (methyl-morphine) is opium alkaloid drug. It has been used as a "gold standard" treatment for cough. Codeine scarcely produce euphoria, and seldom be addicted to. But if used large dose, to some extent, it can produce physical dependence and psychological dependence. There are increasing numbers of cases about its abuse have been reported. But it is still out of control.Liquid phase micro-extraction (LPME) is a novel technique of sample preparation. It is a miniaturized implement of conventional LLE. It is simple, fast, inexpensive, required only micro-liters of organic solvent. In this experiment, we use gas chromatography and hollow fiber liquid phase micro-extraction to determine the content of codeine phosphate in codeine cough syrup.ExperimentalAt first, we prepared HF-LPME device. Then, extracted codeine and its internal standard (Chlorphenamine Maleate) in water by hollow fiber membrane liquid phase micro-extraction, analyzed using GC and found their RT. The second, optimized the option related to extraction. The third, extracted and analyzed codeine in a series of concentration. In the last, calculated and handle the data.ResultsIn the condition of 20℃, 600 r/min stirring speed, adding 200μL,0.01 mol/1 sodium hydroxide, and toluene as organic solvent, we extracted the target chemical(codeine) for 20 min in a series of concentration. There was good linearity at 1.0—50.0μg·mL-1, the average recovery was 102.7%, the relative standard deviation was 4.24%.ConclusionWe have successfully extracted codeine from its cough syrup using new sample preparation method at room temperature (20℃), only 4μL organic solvent was used in each extraction process. Consequently, LPME is a novel, efficient, simple, environment protecting sample preparation. Comparing to other sample preparation, it have such properties: (1) A large choice of extraction solvents can improve selectivity of extraction; (2) The device of extraction is simple and cost is low; (3) Trace sample can be simple, fast, easily extracted, the volume of extract solvent is very small; (4) It can also operate manually and automatically.LPME has a good prospective in the field of pharmaceutical analysis.
