| IntroductionTramadol is a nonopioid analgesics, there are increasing numbers of cases aboutits abuse have been reported. But it is still not be controlled.Liquid phase microextraction(LPME) is a novel technique of sample-preparation.It is a miniaturized implementation of conventional LLE, it is simple, fast, inexpensive,required only microliters of organic solvent.ExperimentalWe at first prepared HF-LPME device.Then, extracted tramadol and pethidine inwater by hollow fiber membrane liquid phase microextration, analysed using GC andfound their RT; The second, optimized the option related to extraction. The third,extracted and analysed tramadol in a series of concetration; Fourly, caculated anddealed with the data.ResultsIn the condition of 20℃,300rpm stirring speed, adding 0.1ml 1 mol/L NaOHsolution, and toluene as organic solvent, we extracted the target chemical(tramadol) for15min in a series concentration of 0.05, 0.10, 0.50, 1.0, 5.0, 10.0μg/ml samplesolutions(water, human urine and human plasma respectively), the RSD of 0.1 and1.0μg/ml: 10.55 and 5.15(water solution);18.06%and 4.03%(urine solution);16.64%and 4.15%(plasma solution).the correlation coefficient attained:0.9997(water),0.9948(urine), 0.9985(plasma). the LODs can reach 0.01μg/ml (water).ConclusionWe have successfully extracted tramaol from biological samples using new samplepreparation method at room temperature(20℃), only 4ul organic solvent was used in each extraction,consequently.LPME is a novel, efficient, simple, environment protecting sample preparation.Comparing to other sample preparation, it have such properties: (1)A large select ofextract solvents can improve selectivity of extraction. (2) The device of extraction issimple and cost is low; (3) Trace sample can be simply, fast, easily extracted, thevolume of extract solvent is very small;(4) It can operate manually and automaticallyalso.LPME has a good prospective in toxicology of forensic science. |
