Objective To investigate the oxidative/anti-oxidative status in SLE-like Model mice and the relationship of oxidation/anti-oxidation indexes,anti-dsDNA and renal involvement.To discuss the effect of lipid oxidation in lupus nephritis and the serum level of anti-dsDNA in SLE-like Model mice.Methods Two groups of(BALB/c×C57BL/6)F1 hybrids were established to SLE-like Mouse Model with treated group(VC+VE) and no treated group respectively.An other group of F1 hybrids was treated as normal control group.Spectrophotometry was used to measure Oxidative/anti-oxidative parameters including malonyldialdehyed(MDA) and total antioxidant capacity(TAOC).The levels of anti-double-stranded DNA (anti-dsDNA) were quantified by enzyme-linked immunosorbent assay(ELISA). Differences of oxidant/antioxidant and autoantibody levels among three groups were analyzed by One-Way ANOVA,relationships between plasma oxidant/antioxidant concentration and anti-dsDNA were tested using pearson correlation analysis.The pathological changes of nephridium were observed by optical microscope and electronmicroscope.Results The levels of MDA(P<0.05,P<0.001,respectively) and anti-dsDNA(P<0.01, P<0.05,respectively) were significantly increased,and the level of TAOC(P<0.01, P<0.001,respectively) was decreased significantly in both treated group and no treated group than that in normal control group.Compared with no treated group,treated group exhibited decreased level of MDA(18.45±3.80 nmol/ml versus 12.99±2.89 nmol/ml, P<0.05) and subsided symptom of renal trauma,but there were no statistically significant differences in the levels of anti-dsDNA and TAOC between the two groups. The levels of MDA was positively correlated with anti-dsDNA(r=0.647,P<0.05),and the levels of TAOC was negatively correlated with anti-dsDNA(r=0.877,P<0.01).The relationship between the levels of MDA and TAOC was negatively correlated(r=0.773, P<0.01).Conclusion Our study found that,SLE-like mice showed exhibited elevated levels of multiple oxidantive markers,which were significantly correlated with anti-dsDNA and renal trauma.The findings suggest that oxidation plays an important role in the pathogenesis and development of SLE.
