Backgroud Holothurian glycosaminoglycan (hGAG) is a kind of polysaccharide extracted from holothurian perisoma. Nowadays, a lot of studies show that polysaccharides, such as plant polysaccharides, fungus polysaccharides, inhibit the growth of the tumor by affecting cell proliferation and inducing cell apoptosis. It has been confirmed that hGAG could have antitumor effect by immunologic mechanism.Objective To investigate the effect of hGAG on the proliferation and apoptosis in human ovarian carcinoma cells SKOV-3 in vitro, and to explore its mechanism of antitumor activity.Methods SKOV-3 cells were cultured in vitro. SKOV-3 cells were dealt with hGAG at different concentration. Cell growth condition was observed by inverted phase contrast microscope. Cell proliferation was analysed by the MTT assay. The cell cycle, apoptotic percentage were detected by flow cytometry (FCM), and the apoptotic cells were detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) in situ.Results The growth state changed when SKOV-3 cells had been treated by hGAG. MTT assay showed that hGAG (0.0001~ 1mg·mL– 1) had inhibited the growth of SKOV-3 cells significantly in a dose dependent manner. When the concentration of hGAG was 1.00mg/mL, the inhibit rate reached to 53.70% at 24 hours. FCM (PI) showed cell cycle had been blocked in G1 phase by hGAG. FCM (AnnexinⅤ/PI) showed that the apoptotic percentage had been 9.55±3.07% by hGAG (1.00mg/mL). The result of TUNEL showed that when the concentration of hGAG was 1.00mg/mL, the apoptosis index could reach to 22.67±4.01%.Conclusion hGAG can inhibit the proliferation of SKOV-3 cells and induce the apoptosis. It may be the one way of its antitumor effects.
