Objective: Tumor is a serious threat to the health of human beings,ovarian cancer is one of the female reproductive system of three kinds of tumor [1],which seriously threat women's health.However,due to ovarian cancer incidence of occult and there is no specific detection index,it become more different for the treatment.In recent years,the reasch of miRNA is the hotspot more and more studies show that miRNA is closely related to the occurrence and development of tumor control,participate in the process of the biological characteristics of tumor cell proliferation,invasion and metastasis,play a similar role of oncogene or anti oncogene [2-4],the study used cell culture,cell transfection,flow cytometry,Real-time PCR(reverse transcription),Western blot(Western blot)expression method of detection of miR-155 and caspase3 in ovarian cancer cell line SKOV3,to investigate the expression of miRNA155 in ovarian cancer cells and its regulating effect on caspase3,and provide a theoretical basis for ovarian cancer gene the treatment means,and provide a new perspective for the diagnosis and treatment of ovarian cancer.Methods:1 Cultivate ovarian carcinoma cell line of SKOV3.2 Group: Though the Hieff Trans liposome transfection reagent,the treatment group was transfected with miR-155 specific overexpress oligonucleotides;the negative control group was transfected with irrelevant miR-155 specific overexpress nucleotide sequence;liposome control group was transfected with Hieff Trans liposome transfection reagent;blank control group no transfection.3 The cellulara growth activity was assayed by MTT.4 Fluorescence activated cell sorting(FACS)was used to analyze the cell apoptosis.5 The miR-155 expression was detected by real-time quantitative RT – PCR.6 The caspase3 protein expression was measured by Western blot analysis.7 Statistical methods: Data were evaluated using spss13.0 statistical software,and date were recorded as mean ± standard deviation(SD).Two groups comparison examined by t test,and means of multiple groups were analyzed using single factor analysis of variance,P<0.05 was considered significant?Results:1 The MTT experiments:0.24.48 hours,the treatment group,the negative control group,blank control group and liposome control group similar growth;72 hours,the growth rate of thetreatment group than other groups of cells grow faster;2 FACS of APC/7-ADD: cell transfection 72 hours later,the treatment group than other groups that early apoptosis rate incress significantly,and has significant differences(P<0.05));the later apoptosis rate have no differents between all the groups(P>0.05).3 Real-time RT-PCR: Compared with Negative control group ? blank control group and liposome control group,treatment group and negative group the miR-155 expression quantity significantly increased,and it has significant difference(P<0.05).4 Western blot: The caspase3 protein expression is obviously higher than that of the negative,blank,and liposome control group,and it has significant difference(P<0.05).Conclusion:1 MiR-155 can inhibit the growth of ovarian cancer cells.2 MiR-155 can inhibit the growth of ovarian cancer cells.3 MiR-155 in ovarian cancer cells with up regulation of caspase3.4 MiR-155 has expression in ovarian cancer cell lines,can regulate the proliferation and apoptosis of ovarian cancer cells through up regulation of caspase3.
