Heart is the largest oxygen-needing organ in body. Heart needs numerous oxygen supplies in time. Blood circulation ensures myocardium acquire sufficient oxygen and nutrition and drain metabolize outcome. The decrease of blood irrigation caused by any reason can bring about ischemia injury to cells. Resuming blood irrigation of tissue as soon as possible is the basic measure to decrease ischemia injury Coronary heart is the most principal disease that affects human death in China. Ischemic heart disease (IHD) is one of the most common cardiovascular diseases in clinical practice. Ischemic heart disease (IHD) is caused by coronal blood circulation changes and induces the imbalance of myocardial supply and demand. And its pathogenesis is complex. Drug therapy is one of important ways to cure myocardial ischemia. Although nitrate, calcium-channel blocker andβ-receptor antagonist are effective in the therapy of myocardial ischemia, they all have some adverse reactions. Studies on the new agents used to treat myocardium ischemia are still a pivotal issue. Recent studies have showed that oxidative stress is one of the important reasons to cause coronary heart.Tribulus terrestris is also called Bai Jili and Ci Jili. It refers to the dry, ripe fruit of the plant caltrop. Its main chemical ingredients are saponins, ketone, alkaloid. The gross saponin of Tribulus terrestris(GSTT) was one of the effective group ingredients which was exracted from the whole plant of caltrop. It is a mixture of over 10 kinds of saponins, mainly steroid saponins. It mainly includes furan-strol and spiral-strol. It is a powder and dissolvable in water. Domestic research materials showed that GSTT had: the effect of lowering blood pressure and stimulating anti-hypoxia, sheeting blood vessels, lowering cholesterol, inhibiting fat deposition on arteries, myocardium and liver, anti-aging property and invigorating sexual function. Capsuled praeparatum of GSTT has been utilized for the diseases of cordis and cerebral vascular system generally in clinical treatment with curative effect obviously. However, how GSTT protects myocardium cell against damage during hydrogen peroxide and what is the mechanism need further investigate. the objectives of the present study are to copy the model of myocardial ischemia in vitro to invesgate the protective effects and possible mechanisms of gross saponins of Tribulus terrestris on the cardiovascular disease and to provide a theoretical basis for better, safer use and targeting application in clinical practice.The primary cultured cardiac myocytes in neonatal rats damaged by 0.3 mmol·L-1 hydrogen peroside (H2O2) simulated the oxidative stress model. The survival ability was detected by Methyl thiazolyl tetrazolium (MTT) method. The content of LDH in cultured conditions, SOD,CK and MDA in myocytes were measured by the instruments. Cell apoptosis rate and the the influence of the intracellular content of NF-κB were detected by flowcytomety. The apoptosis cells morphous were observed by hoechst33258 through fluorescence microscope, NF-κB were analyzed by Western blot.The results experiments showed that numerous fragmentation and crimple of cardiocyte after incubation with 0.3 mmol·L-1 H2O2, cell swelled and adherence cell quantity reduced, nucleus was stained deeply and concentrated. GSTT could increase the survival rate of cardiac myocytes, reduce apoptosis percentage, up-regulated NF-κB expressions, reduce LDH, CK leakage, MDA decreased and and stimulates the SOD activeness to increase, And GSTT can reinforce NF-κB translocatefrom cytoplasm to cellular nucleus. Chelerythine abolished these effects partly.In conclusion, gross saponin of tribulus terrestrise reducing the the generation of MDA and increasing the activity of SOD inmyocytes damaged by H2O2,reduce LDH, CK leakage,reduce free radicals on myocardium cells damage. Indirectly demonstrated that gross saponin of tribulus terrestris could protect the activities of antioxidase and inhibit the formation of lipid. Also hinted that one of probably mechanisms of gross saponin of tribulus terrestris on protecting myocardium is inhibiting the production of oxygen free radicals. Our study demonstrated gross saponins of Tribulus terrestris can effectively inhibit cadiocyte apoptosis induced by H2O2,the study showed in the GSTT group NF-κB content was significantly higher than Chelerythrine; Chelerythrine inhibited NF-κB content; Protective effect of gross saponin of tribulus terrestris on myocardial cells against H2O2injury protection may be through activating PKC- NF-κB signal transduction pathway.
