Objective : To study the relationship between expression of PTEN , VEGF and the behavior of clinical pathobiology as well as their associativity. Methods: To select randomly paraffin block specimen at the China-Japan Union Hospital of Jilin University and chang chun Obstetrics and Gynecology hospital in 2004 Feb-2008 Feb ,including 20 cases of normal endometrial tissues,20 cases of hyperplasia tissues of endometrium (including 8 cases of simple type and 12 cases of complicated type),10 cases of atypical hyperplasia tissues and 42 cases of endometrical cacinoma tissues (which were divided according to pathologicaltypes,clinicalstage,differentiation,the depth of infiltration in muscular layer and metastatic of lymph node) .All the tissues were treated without adjuvant radiotherapy , chemotherapy and hormonal therapy before operation。These tissues were stained by immunohisto- chemistry SP method to detect the expression of PTEN and VEGF .Result:①The positive rates of PTEN and VEGF were 100.00%,10.00% in normal endometrial tissues , 95.00%,20.00% in hyperplasia tissues of endometrium , 60.00%,60.00% in atypical hyperplasia tissues and 42.86 %,71.43% in endometrical cacinoma tissues. Compared with groups of normal endometrial tissues and hyperplasia tissues of endometrium,the positive rates of PTEN and VEGF in endometrical cacinoma tissues were significantly different (p<0.05,P<0.01).The expression of PTEN and VEGF were correlated with development of endometrical carcinoma.②The positive rates of PTEN in endometrical carcinoma of well /moderately differentiation and poorly differentiation were 53.13%,10.00%,two groups were significantly different (P<0.05). The positive rates of VEGF in endometrical carcinoma of well /moderately differentiation and poorly differentiation were 62.50%,100.00%,two groups were significantly different (P<0.05).③The positive rates of PTEN in the group of muscular layer without infiltration and the depth of infiltration in muscular layer achieved to≤1 /2 and >1/2 were 59.26%,13.33%.There was significant difference between two groups statistically (p<0.05) . The positive rates of VEGF in the group of muscular layer without infiltration and the depth of infiltration in muscular layer achieved to≤1 /2 and >1/2 were 59.26%,93.33 % .There was significant difference between two groups statistically(p<0.05) .④The positive rates of PTEN in the group of metastases of lymph node and metastatic of lymph node were 50.00%,12.50%.There was no significant difference between two groups statistically (p>0.05) . The positive rates of VEGF in the group of metastases of lymph node and metastatic of lymph node were 64.71%,100.00%.There was no significant difference between two groups statistically(p>0.05) .⑤The positive expression of PTEN in adenocarcinoma endometrium and squamous carcinoma were 41,67%,50.00%, there was no significant difference between two groups statistically (p>0.05). The positive expression of VEGF in adenocarcinoma endometrium and squamous carcinoma were 69.44%,83.33%, there was no significant difference between two groups statistically (p>0.05).⑥There was a nagative correlation between the positive rates of PTEN and the positive rates of VEGF. (γ=-0.633,P<0.05)。Conclusions:1.Normal expression of PTEN was one of the critical regulatory agents in normal differentiation of intimal epithelial cells.Failure in expression of PTEN encourged the development of endometrical carcinoma。2. Hyperexpression of VEGF encourged the development of endometrical carcinoma.3.Positive rates of PTEN were negatively correlated with histo- differentiation and oncoma invasion . The more lower positive rates of PTEN , the more higher malignant degree of the carcinoma。4.Positive rates of VEGF were positively correlated with histo- differentiation , oncoma invasion and metastatic lymph node.5. PTEN and VEGF were nagatively correlated with the development of endometrical carcinoma.6. We can judge malignant degree of the tumor on monitoring the expression of PTEN and VEGF.
