Objective:In the present study, we investigated into the activity and mechanism of Huanglian jiedu decoction(HJD) against parkinson's disease in vivo and in vitro.Methods: Using C57/BL mice as the vivo demage model of parkinson's disease, 25mg·kg-1 MPTP were injected peritoneal. The effect of HJD on behavioral test of mice model was observed that hypokinesia was alleciated that the time in pole test decreased, the traction accrete increased, and locomotor coordination in swim test improved. Activities of SOD,MDA and GSH-PX in the brain were detected. Cell model of Parkinson's disease was established by adding MPP+ into PC12 cell. The protective effects of rat serum concentrating HJD or its components on PC12 cell were assessed. The cell viability was analyzed by methyl thiazolyl tetrazolium(MTT) method, the morphological changes were observed by acridine orange (AO) and ethidium bromide (EB) staining assay. Cell cycle and mitochondrial transmembrane potential were measured by flow cytometry.Results: The effect of HJD on behavioral test of parkinson's disease mice was observed that hypokinesia was alleciated that the time in pole test was shortened(P<0.05), the traction accrete increased (P<0.05), and locomotor coordination in swim test improved compared to control group. SOD activity in brain was stimulated and MDA expressing level was promoted by HJD. Viability of PC12 cell injury induced by MPP+ was significant enhanced by the serum concentrating HJD(P <0.01). Furthermore, the serum concentrating gardenia, also contribute to the viability of PC12 cell (P<0.05). the serum concentrating Scutellariae and gardenia can protect PC12 cell, but the effect was not as strong as complete and gardenia. In the baicalein,baicalin,Wogonin,wogonoside and jasminoidin, baicalein showed dose-dependent in protection of PC12, while the other four components'effect was not obevious. Under 200μmol?l-1 MPP+, the morphological of PC12 cell was changed, including apoptosis and necrosis. The serum concentrating HJD can notable protect these injuries of PC12 cell. Using flow cytometry, we can observe the serum concentrating HJD can decrease the percentage of G1 phase and increase the percentage of S phase and G2 phase. The protection on the mitochondrial membrane potential of PC12 was also observed.Conclusions: 1 intrperitoneal MPTP 25mg·kg-1 for 8 days there is a series of ethology change to be similar to parkinson's disease ,as body thrill and muscle stiffness. to observe ethology change of mice,we make sure that the Parkinson's disease mode induced by MPTP is succeed.2 Huanglian jiedu decoction may obviously improve ethology change and depress the level of SOD and MDA of mice brain tissue, show that Huanglian jiedu decoction have the prevent effect on the whole level to MPTP Parkinson mode.3 the blood serum of Huanglian jiedu decoction have the conspicuous protect effect to the damaged PC12 by MPP+, gardenia have the same effect. Scutellaria and gardenia also have the protect effect but is weaker than complete and gardenia。The major active component of Huanglian jiedu decoction come from gardenia and scutellaria, coptis and phellodendr may have the cooperation .4 accoding to compared with the five element in Huanglian jiedu decoction , baicalein have the powest effect of protect cell archaeus ,it shows that baicalein is the active component of Huanglianjiedu decoction prevent and treat parkinson's disease.5 MPP+ induce PC12 cell cycle changed and depressed mitochondria membrane potential. blood serum of Huanglian jiedu can decrease the change in some extent, backswing cell cycle to normal direction and increase mitochondria membrane potential.supposed that Huanglian jiedu decoction protect PC12 by regulation cell cycle and mitochondria membrane potential.
