Objective: To investigate the expression and correlation of Skp2 and P27kip1 in colon carcinoma and detect the relationship with the development of the diseases.Method: To detect the protein expression of Skp2 and P27kip1 by imrnunohistochemistry staining (SABC method) in 50 cases of colon cancer and 20 normal colonic mucosas. The result, which estimate according to the intension of stain, Was classified into 0-3 scores. 0 (no colored), 1 score (slight), 2 scores (moderate), 3 scores (strength). The stain extent, which according to the percent of positive cells, was divided into 0-3 scores. 0 (0%), 1 (<25%), 2(25-50%), 3 (>50%) . The final scores were obtained by adding extent of stain score and intension of stain score together. The final score which was less than 2 was negative cases, the positive cases was more the 2. statistical analysis made by spss11.0 software according to clinicopathological data of colon cancer.Result: the positive expression of Skp2 and P27kip1 mainly located within the cellular nucleus. The protein expression of skp2 was 64.0%, significantly higher than normal colonic mucosa. 15.0% (P<0.05). The protein expression of P27kip1 was 22.0% in the colonic cancer, significantly lower than normal colonic mucosa. 90.0% (P<0.05). Both expression of Skp2 and P27kip1 have negative relationship with age and sex. But they have the positive relationship with the differentiation, Dukes staging and lymph node metastases (P<0.05). The negative correlation was present between the Skp2 and P27kip1( r=-0.282, P<0.05).The positive expression of p53 mainly located within the cellular nucleus and was brown. There is no expression in the normal colon tissue (0.0%, 0/20) but there was significant increased expression in the colon cancer (80.0%, 40/50, P<0.01) . The positive express of the bcl-2 gene , which located within cytoplasm, can be detected both in the normal colon and colon cancer. There was a significant increase of positive expression of bcl-2 in the colon cancer 74.0% (37/50) compared with the normal tissue 10.0% (2/20) (P<0.05). The expressions of both p53 and bcl-2 have the significant relationship with the pathologic differentiation of colon cancer, but have no relationship with age, sex, stage and lymph metastasis. Skp2 has positive correlation with p53, bcl-2 respectively(r =0.345, r=0.231,P<0.01). Conclusion: The abnormal expression of both Skp2 and P27kip1 is one of the reasons that cause the development of colon cancer. It may be realized through the up-regulate the expression of p53 and bcl-2. The abnormal expression of Skp2 and P27kip1 has special relationship with the differentiation, Dukes staging and lymph node metastases of colon cancer respectively.
