Objective: To investigate therapeutic effect of monomer Rg1, Rb1 of Panax notoginseng saponins(PNS) on liver fibrosis of experimental rats.Methods: 75 healthy Sprague-Dawley(SD) rats with 180±20g, and half of male and female each, were bought from The Experimental animal Center of Kunming Medical College. The animals were randomly divided into 5 groups: group 1(Rg1 60mg/kg/d), group 2(Rb1 60mg/kg/d), group 3(PNS 60mg/kg/d), group 4(model) and group 5(normal control). Each group had 15 rats. All groups were given CCL4 by intradermic to establish the model of liver fibrosis except group 5 injected with same dosage olive oil. At the same time, the rats of group 4 were injected saline(1ml/d) by intradermic; the others were given drugs once a day(the dosage as mentioned above). In the first week of injection, one rat was sacrificed randomly in each group; in the second and third week, two rats(one male and female) were sacrificed randomly. The liver of rats was taken in order to observe the process of liver fibrosis. During the experiment period, the general condition, behavior and weight change of rats were observed. According to the pathological changes, the degree of liver fibrosis and inflammation were divided into 5 grades with HE staining, Masson's staining and Gomori's staining. In addition, blood of rats was dropped to measure liver function such as ALT, AST, TB, ALP and serological index of liver fibrosis including LN, PCIII, HA.Results:①During 5 weeks, 3 rats of group 2 and 1 of group 3 died. Rats of other groups are all alive. All the rats except the 4 were all alive. The general condition of rats of group 2 was worse than that of group 5. The general condition of rats of group 1, group 3 was better than that of group 4 and group 2.②The bodyweight of all groups was different after experiment (P<0.05), the changes of bodyweight had a significant statistical difference in all groups(P<0.01); the bodyweight of the group 2, group 3 and group 4 was observed different from group 5(P<0.01); between the group 1 and group 5 there was no statistical difference; between group 1 and group 2 had difference(P<0.05).③The stages of liver fibrosis among all groups had significant statistical difference(P<0.01); The inflammation degree among all groups also had statistical different(P<0.05).④ALT, AST, TB, ALP and LN, PCIII, HA among all groups was different(P<0.05). These of group 2 were observed higher than group 5 significantly. These of group 3, group 1 were observed lower than group 4 and group 2.Conclusion:1. The rat model of liver fibrosis induced by CCL4 is a simple, easy and cheaper experimental model.2. The Rb1 have toxic effects on liver tissue without any therapeutic effect for liver fibrosis of rats. The antifibrotic effect of Rg1 on animal model was more obvious than that of PNS, with less toxic effect than Rb1. Therefore, Rg1 could be regarded as a reasonable traditional Chinese antifibrotic agent.
