【Objective】To investigate the significance and the clinical value of the expression of PTEN ,Bcl-2 in osteosarcoma .【Methods】mmunohistochemistry S-P od was used to detect PTEN ,Bcl-2 in 62 cases of osteosarcoma(50 cases of primary tumors and 12 cases of recurrent tumors, 15 cases with lung metastasis and 47cases without lung metastasis).The relationship among PTEN or Bcl-2 expression, various clinicopathological factors and the prognosis of osteosarcoma was analysed. The relationship between the expression of PTEN and Bcl-2 was investigated as well.According to the rate of the positive tumor cells, the results of PTEN and Bcl-2 were semi-quantitated.【Results】1. The expression of PTEN in osteosarcoma and its clinical significance. (1)Normal tissue was positive for PTEN, but a relatively low rate was found in osteosarcoma .Of the 62 patients with osteosarcoma, 36(58.1%) showed positive staining of PTEN in the primary lesion. (2)In osteosarcoma,The expression of PTEN protein was related with Lung metastasis and survivaltime (P<0.05) . (3) In osteosarcoma,The expression of PTEN protein was not related with gender,Tissue types,Differentiation (P>0.05 ) . (4) The positive rate of PTEN was 58.1 %( 36/62) in osteosarcoma group and 85 %( 17/20) in osteochondroma group.There was a prominent difference between them (p<0.05).2. The expression of BCL-2 in osteosarcoma and its clinical significance. (1)Normal tissue was almost negative for BCL-2, but are latively high rate was found in osteosarcoma.Of the 62 patients with osteosarcoma, 32 (51.6%) were expressed positively for BCL-2. (2)In osteosarcoma , The expression of BCL-2 protein was related with differentiation (P<0.05) . (3) In osteosarcoma,The expression of BCL-2 protein was not related with gender,Tissue types,Lung metastasis,survivaltime (P>0.05 ) . (4) The positive rate of BCL-2 was 51.6 %( 32/62) in osteosarcoma group and 20 %( 4/20) in osteochondroma group.There was a prominent difference between them (p<0.05).【Conclusion】These data suggest the PTEN expression correlate with BCL-2 expression and both may contribute to the genenis and progression of osteochondroma .There existed low expression of PTEN and over-expression Bcl-2 proteins .Coexpression of these antioncogenes played an important role in invasion and rnatastasis in osteosarcoma.
