| Objective To explore the relationship between Toll-like receptors(TLRs) and prognosis of severe viral hapatitsis by examing the change of Toll-like-receptor expression and the level of serum endotoxin.Methods 47 patients of severe viral hapatitsis were divided into death group and survival group,meanwhile 12 health volunteers were served as control .The peripheral blood mononuclear cells were separated and the expression of TLR2 and TLR4 mRNA was observed by method of RT-PCR,The level of serum endotoxin was measured by chromogenic substrate Limulus amebocyte lysate assay.Results The expression of TLR2 and TLR4 mRNA and the level of serum endotoxin of death group before therapy were lightly higher than that of survival group,but after therapy that of death group is higher than survival group(p<0.05).and they all had significant difference comparing with control group(p<0.01). the expression of TLR2 and TLR4 mRNA were positively correlated with the level of serum endotoxin.Conclusions TLR2,TLR4 and serum endotoxin have important role in severe viral hapatitsis and the change of TLR2 and TLR4 mRNA were closely correlated with the level of serum endotoxin.They might predict fairly the trend of severe hepatitis prognosis and can be used as indices of evaluating severe hepatitis prognosis. |
