Guided bone regeneration (GBR) methods have been widely used in implant supported prosthodontics. Chosing good GBR membrane is essential to the success of GBR methods, so the research of GBR membrane is the hot spot now. This study examined the attatachment and proliferation of MG63 osteoblast-like cells on Ag-nHA-nTiO2/polyamide-66 nanocomposite membrane for understanding the biocompatible of the membrane in some degree.The Ag-nHA-nTiO2/PA66 membrane was experimental group, nHA/PA66 membrane and expanded polytetrafluroethylene membrane were control groups in this study. Light microscope and scanning electronic microscope were used to observe the microstructure of the three kinds of membrane.MG63 osteoblast-like cells were cultured on the three kinds of membrane and tissue culture polystyrene (TCPS).The cell proliferation was checked by MTT method, the cell viability was checked by Cell Viability Analyzer, Alkaline phosphatase activity and Ca2+ concentration of osteoblast-like cell matrix were detected by ELISA. Scanning electronic microscope was used to observe the adhesion and proliferation of the MG63 osteoblast-like cells on the three kinds of membrane.SEM showed that the Ag-nHA-nTiO2/PA66 membrane is composed of the obverse face and the opposite face.The obverse face is porous and the opposite face was smooth.The shapes of pores of the obvers face are different and the diameters of those are from 50um to 300um.The opposite face showed some tiny pores(5-10um in diameters).The microstucture of nHA/PA66 membrane is the same as the Ag-nHA-nTiO2/PA66 membrane.microstuctures of the obverse and the opporsite face of the e-PTFE membrane are same. The e-PTFE membrane showed many tiny lined cracks (about 3-5um in width and 15um in length) in elliptic structure. The transeverse section of the Ag-nHA-nTiO2/PA66 membrane showed many macropoles of the obvers side are connected with each other and some tiny pores of the opporsite side are neither.The same to nHA/PA66 membrane.The transeverse section of the e-PTFE membrane showed the crackes are connected with each other from the obverse side to the opporsite side. MTT assay showed that there no significant difference in the three kinds of membrane (P>0.05).The cell viability of the MG63 osteoblast-like cells on the Ag-nHA-nTiO2/PA66 membrane was lower than the control groups (P<0.05).The alkaline phosphatase activity and the Ca2+ concentration of osteoblast-like cell matrix showed that there no significant difference in the three kinds of membrane groups and the TCPS (P>0.05). The adhesion and proliferation of osteoblast-like cells on the three kinks of membrane were both well.The adhesion of osteoblast-like cells on the Ag-nHA-nTiO2/PA66 membrane are better than the e-PTFE membrane.Based on these findings, the Ag-nHA-nTiO2/PA66 membrane has no negative effects on the growth of osteoblast-like cells.Because of good biocompatibility and the graded stucture of the Ag-nHA-nTiO2/PA66 membrane, it could be better than the e-PTFE membrane.
