Objective: To investigate the expression of phosphoinositide 3-kinase(PI3K) and protein kinase B(Akt) and their relation in pathogenesis in laryngral squamous cell carcinoma.Methods: Immunohistochemical S-P method was used to detect the expression of PI3K and p-Akt in 40 paraffin-embedded human laryngral and 20 cases of polyp of vocal cords and 20 cases of laryngeal papilloma. All the samples were stained by HE method to confirm their pathologic characters. Positivity rates of PI3K and p-Akt were evaluated by microscop.Results1. The positive rates of PI3K in laryngeal squamous carcinoma and polyp of vocal cord were 82.5%(33/40) and 15% respectively, the positive rates of p-Akt in laryngeal squamous carcinoma and polyp of vocal cord were 92.5%(37/40) and 10% respectively. The difference of the PI3K and p-Akt expression between the experimental group and control group was significant (p<0.01).2. The positive rates of PI3K in laryngeal papilloma were 75%(15/20) , the difference of the PI3K expression between the laryngeal squamous carcinoma group and laryngeal papilloma group was quiet(p>0.05);the positive rates of p-Akt in laryngeal papilloma were 20%(4/20),The difference of the p-Akt expression between the laryngeal squamous carcinoma group and laryngeal papilloma group was significant (p<0.01).3. PI3K and p-Akt expression in patients with pathological grading (stagesⅠandⅡ) were significantly higher than in other patients with pathological grading (Ⅲ)( p<0.01).A positive correlation was find between PI3K and p-Akt in laryngeal squamous cell carcinoma.Conclusions1. The up-regulated expression of PI3K and p-Akt in laryngeal squamous carcinoma suggests that the abnormal activation of PI3K/Akt pathway is tight related with the tumor occurrence and progress.2. PI3K and p-Akt expression in patients with pathological grading (stagesⅠandⅡ) were significantly higher than in other patients with pathological grading (Ⅲ)。The abnormal expression of PI3K and p-Akt in laryngeal squamous carcinoma suggests their important roles in the oncogenesis and cell differentiation . The immunohistochemistry dyeing of PI3K and p-Akt is also a morphology factor for tumor cell differentiation.
