Objective: Intracerebral hemorrhage which is a clinical disease usually be found in old persons,even it can endanger their life.Edema in brain parenchyma surrounding an intracerebral hemorrhage aroses incereased intacranial pressure which usually brings on death or deformity of patients. The mechanism of brain edema surrounding an intracerebral hemorrhage is complicated. Hemoglbin,one of the complications which are included in the brain edema after ICH gets far-ranging attention.We tried to act on rats injected hemoglbin. The brain water content , the expression of AQP4 mRNA and AQP4 protein could be observed that they are influenced by HB.To research the effect of hemoglobin in the edema in brain parenchyma injection hemoglobin,and to provide the new usage of treatment about brain edema.Method: 160 rats were divided into control group(n=10),experimental group(n=150). Experimental models were built on the rats in experimental group.And then,these rats were divided into model group,AQP4 intervenor group,HO-1 intervenor group randomly.Animals in intervenor groups were irrigated allyl chloride (200mg/kg)into stomach,or injected Znpp(45umol/kg)into peritoneums once a day. We graded the neurological function, observed the brain samples under microscope, and examined the brain water content and expression of AQP4 mRNA and AQP4 protein by methods of RT-PCR and western blot at the different time(3h,12h,1d,3d,5d).And then ,we analysed the changes and the connection between them.Result: (1) The neurological functional score of rats in control group behind operation was not higher than that before operation. But the neurological functional score of rats in model group behind operation was evidently higher than that before operation. At the 12h to 24h after operation ,the neurological functional scores were highest. (2) The brain water content of model group raised evidently compared with control group.At the first day after operation, the brain water content was the greatest. Comparison the brain water content of two sides of model group,the side of injection hemoglobin was greater than the anthor side.The brain water content was statistic difference between model group and intervenor groups.But the brain water content of intervenor groups was evidently greater than that of control group . There was no remarkable difference between the brain water content of AQP4 intervenor group and HO-1 intervenor group. (3) Observing the brain samples under microscope we could not find abnormity in brain organise and configuration in control group.Both the different time, specially 1d after operation, cells edema and destroy of cells'configuration in brain organise in model group.But cells edema and destroy of cells'configuration in brain organise in intervenor groups were statistic lighter than that of model group. (4) The quantity of AQP4 mRNA was measured by methods of RT-PCR.We observed it in model group was statisticsly higher than that of control group.The quantity of AQP4 mRNA in intervenor groups was remarkably lower than that in model group.Yet, the quantity of AQP4 mRNA in intervenor groups obviously increased compared with control group.(5) Change of expression of AQP4 protein was the same like the change of expression of AQP4 mRNA. We observed it in model group was statisticsly higher than that of control group. The quantity of AQP4 protein in intervenor groups was remarkably lower than that in model group.But the quantity of AQP4 protein in intervenor groups obviously increased compared with control group.Conclusion: (1) Injection HB into the caudate nucleus of rats could bring on brain edema successfully. (2) Giving allyl chloride could reduce Brain edema and abnormity in brain organise and configuration which could be induce by injection HB. (3) Giving HO-1 inhibitor (Zn-protoporphyrin) could reduce Brain edema and abnormity in brain organize and configuration which could be induce by injection HB.The effect suggested that AQP4 be concerned with brain edema's forming of Pathophysiolog process induced by HB at first hand.ZnPP and allyl chloride can regulate expression and function of AQP4 to restrain brain edema induced by HB.
