With the development of globalization in economy and regional peace, and the progressing improvement of living standard and the variation of life style , drugs abuse has overrun around the whole world, giving rise to more and more serious social and medical issues. Then the prohibition of illegal traffic, of preparing and of selling drugs, and therapy to addiction would be becoming a hot issue. The study of mechanism on drug dependence and new medication to detoxification are flourishing.Up to now, the principal accomplishment in managing of the physical dependence have been achieved, however, people have not found out the mystery of psychical dependence, being incapable of preventing relapse. Moreover, there is not a model of relapse of psychical dependence acknowledged by the public at present.CART (Cocaine and Amphetamine Regulated Transcript) peptides may be a new neurotransmitters involved in reward and reinforcement, feeding and satiety, stress, endocrine regulation, sensory processing and other physiological processes. CART mRNA was increased in rat striatum after acute administration of cocaine or amphetamine. We used methods were establish morphine dependence model of pregnant rat, and then made E10,E11,E12,E13,E14,E16,E18,E20 and P0 nine times morphine dependence and normal sample of fetus rat, at last, the ontogeny of cocaine- and amphetamine-regulated transcript (CART) expression has been analyzed by immunohistochemistry in the hypothalamus of the rat and compared with the Rat of dependence on morphine. The main results are as follows:1.The numbers of CART-ir neurons in the anterior, middle and posterior hypothalamus of fetus rats have an increase by degrees tendency during prior to birth , but decrease in postnatal.2.The numbers of CART-ir neurons in the anterior, middle and posterior hypothalamus of the morphine dependence fetus rats have an increase by degrees tendency during development.3.CART-like-immunoactivety (CARTLI) neurons were first detected on the embryonic day 16 (E16) in the hypothalamus of the normal rat , and E14 in the hypothalamus of the morphine dependence rat.4.The ontogeny of CART expression in the hypothalamus of the morphine dependence rat are more then the normal fetus rat. With all the above results, we conclude that:1.The numbers of CART-ir neurons in the hypothalamus of fetus rats have an increase by degrees tendency during prior to birth , but decrease in postnatal.2 . Chronicity morphine treatment with rat advanced CART-ir neurons in the hypothalamus expressed increase and early.3.CART-ir neurons in the hypothalamus expressed increase after withdrawal maybe concerned with stress.
