Injury Actions Of Ultrasound-guided Percutaneous Ethanol Injection On Rabbit Liver

Objective:To explore the effect of Percutaneous ethanol injection(PEI) therapy on liver,and whether this action is in relationship with dosage,time, and position,in order to provid an experimental base for more scientific applying of PEI in clinical service.Methods:16 ordinary rabbits were divided into two groups,respectively received intrahepatic injection of 1ml,2ml absolute alcohol under ultrasonic guidance.Another 4 ordinary rabbits were in control group.All animals were detected in numbers per unit area of blood vessel in injection region,the level of AST and ALT,oxidation-deoxidize enzyme,alcohol metabolic enzyme, apoptotic index(AI),by methods of ultrasonic image,metabolize enzyme, immunohistory,and TUNEL.Results:The levels of AST,ALT,MDA in experimental rabbits were significantly higher than those in control rabbits(P＜0.05),and these changes had positive dosage-dependence effect of injected alcohol,while decreased by time;The content of SOD and GSH in experimental group decreased notably, compared with that in control group after the first day of PEI,But then increased by time,and the contents of GSH in different group had statistical difference with each other(P＜0.05).The enzymatic activity of CYP2E1 increased with the addition of alcohol injection dosage(P＜0.05),and the activity of CYP2E1 around 1cm from the injection point was much higher than that around both Ocm and Xcm(P＜0.05).The apoptosis correlated with the injection dose and position. Apoptotic index of experimental rabbits increased with the rise of alcohol dose, which was significantly different from that of control rabbits.The rate of apoptosis was lowest at the injection point,while it was highest around 1cm from the injection point,and the difference is statistically significant (P＜0.05).Moreover,the ultrasonic image showed that numbers per unit area of blood vessel in injection region of the rabbits were fewer after received injection of 2ml alcohol(P＜0.01)Conclusions:PEI do harm to the liver of rabbit in some extent,The damage has positive dosage-effect relationship with injected alcohol,and correlates to time and position.
