Objective:To evaluate the expression,variance and clinical significance of the tumor suppress gene WWOX in hepatocellular carcinoma (HCC), para-carcinoma tissue,normal liver tissue and hepatoma cell lines (HepG2 and SMMC-7721 ) by RT-PCR and FQ-PCR.Methods:①The specific primers and TaqMan probe of WWOX were designed according to foreign document. and WWOX mRNA (AF211943) is searched from GENEBANK by bioinformatics and its realities were verified by primer express 2.0(a TaqMan probe designing software) and primer premier 5.0.②The total RNA isolated from 40 pairs of human HCC tissue and para-carcinoma tissue, normal broken liver tissue and two hepatoma cell lines (HepG2 and SMMC-7721 ), was reversely transcribed into cDNA.③Housekeeping gene GAPDH was used as internal reference. The RT-PCR method was used to analyze the variance of WWOX gene.④Housekeeping geneβ-globin was used as internal reference. The real-time fluorescence quantitative PCR method was used to analyze the expression level of WWOX gene.Results:①Expression of WWOX mRNA in hepatoma cell lines SMMC-7721 which is significantly less than that in another hepatoma cell lines HepG2 is relactively close to that in HCC tissue. ②Expression of WWOX mRNA in HCC(0.318±0.0559)is significantly less than that in para-carcinoma tissue(1.0),P<0.05.③Positive percentages of WWOX in human HCC, para-carcinoma tissue, normal broken liver tissue are 52.5%,77.5%,100%,respectively,which are different significantly ,p﹤0.05. Percentage of human HCC(55%) is significantly lower than that in normal broken liver tissue(100%),P﹤0.05. That in para-carcinoma tissue (77.5%)is no lower than that in normal broken liver tissue(100%),P﹥0.05.④Variety ?6-8 in HCC, and its frequency 20%(8/40) is significantly higher than that in para-carcinoma tissue 2.5%(1/40) ,P<0.05.⑤Expression of WWOX were significantly correlated with pathological grades, tumor thrombus and HBV status in serum(P<0.05). In contrast, it was not associated with gender,age,tumor size,lymph node metastasis or tumor encapsulation(P>0.05).Conclusions:①WWOX gene is an tumor suppress gene .The decreased expression and variance of WWOX gene may play an important role in the carcinogenesis and development of HCC. WWOX gene may become a new target site for gene therapy of HCC in future.②Action of WWOX protain by hormone or hormone receptor maybe one of its functions.③The expression of WWOX mRNA in HCC was related to pathological grades and tumor thrombus in serum, which indicates WWOX expression maybe the reference guideline for prognosis and malignant degree in HCC.④The expression of WWOX mRNA in HCC was related to HBV status in serum, which indicates WWOX maybe one of paths that HBV cause HCC in. |
