Objective:To determine the depression effect of cell proliferation and synthesis of collagen when 5-fluorouracil interfere in the fibroblasts of pathologic scars .Methods:Fibroblasts were derived from four hypertrophic scars,four keloids ,four normal skins ;Cell culture was enforced by the method of literature .Transfer of culture was operated when cell fused.4-6 generation cell was use in the experiment .Cell viability after interfered by 5-fluorouracil for different minutes (1,2,5,10,15,30min)was evaluated by MTT assay compared with the control group ;After get the optimalizing interfered time ,we use different doses of 5-FU interfere in the fibroblasts deviced from three different skins .The technology of incorporation of 3H-proline was used for detecting the capablitity of fibroblasts secrete ,comparing with that hasn't interfered.Gene expression of collagen typesâ… andâ…¢were quantified by means of reverse-transcriptase polymerase chain reaction assays.Results:Sharp of cell kept normal after interferd by 5-FU,The depression effect of fibroblasts interfered by 5-fluorouracil for 10 min was significant comparing with the control and the team interfered for 5 min ,but not significant comparing with the team interfered for 30min ,so was the synthesis of collagen. And the depression effect of fibroblasts interfered by 5-fluorouracil in the dose of 25mg/ml for 10min was significant comparing with the control and the dose of 12.5mg/ml ,but was not significant comparing with the team of dose of 50mg/ml ,so was the synthesis of collagen. The gene expression of collagen typesâ… andâ…¢had no significant difference.Conclusions: 5-fluorouracil on the concentrationcan of 25mg/ml depress the capablitity of fibroblasts secrete collagen protein after interfering in them for 10 min ,but collagen typesâ… andâ…¢mRNA were unaffected by 5-FU treatment.
