Objective This experiment is to investigate effects of drugs as 1,25-hydroxyvitamin D3, Methotrexate and disodium cromoglycate, on proliferation and collagen synthesis of hypertrophic scar fibroblasts in vitro, and to evaluate the possibility of drugs in the prevention and treatment of hypertrophic scar. Methods The primary cultured fibroblasts derived from resected human hypertrophic scar were used to establish the monolayer culture model. cells of passage 48 were cultured with media which contain 1,25-hydroxyvitamin D3 (10-11mol/L, 10-10mol/L, 10-9mol/L) , Methotrexate (10-9mol/L, 10-8mol/L, 10-7mol/L, 10-6mol/L, 10-5mol/L) and Disodium cromoglycate (10-9mol/L, 10-8mol/L, 10-7mol/L, 10-6mol/L) , MTT assay and Hydroxyproline test were used to detect the effects of different drugs on the proliferation and collagen synthesis of hypertrophic scar fibroblasts, LDH test was used to detect drugs' cytotoxicity.Result 1, 1,25-hydroxyvitamin D3 significantly inhibited proliferation hypertrophic scar fibroblasts with 10-10mol/L and 10-9mol/L doses, compared with ethyl alcohol control group(P<0.01); Methotrexate with 10-6mol/L and 10-5mol/L doses and Disodium cromoglycate with 10-6mol/L dose significantly inhibited proliferation hypertrophic scar fibroblasts with 10-10mol/L and 10-9mol/L doses, compared with control group(P<0.01). 2, Methotrexate with 10-7mol/L10-5mol/L doses had cytotoxicity on hypertrophic scar fibroblasts(P<0.05); While 1,25-hydroxyvitamin D3 with 10-11mol/L10-9mol/L doses and Disodium cromoglycate with 10-9mol/L 10-6mol/L doses had no cytotoxicity on hypertrophic scar fibroblasts(P<0.05). 3, 1,25-hydroxyvitamin D3 with 10-11mol/L10-9mol/L doses inhibited collagen synthesis of hypertrophic scar fibroblasts, compared with ethyl alcohol control group(P<0.01), Methotrexate with 10-9mol/L10-5mol/L doses and Disodium cromoglycate with 10-6mol/L dose inhibited collagen synthesis of hypertrophic scar fibroblasts, compared with control group (P<0.01) . Conclusion 1, 1,25-hydroxyvitamin D3 and Methotrexate can significantly inhibit the proliferation and collagen synthesis of hypertrophic scar fibroblasts in vitro, which may be effective on prevention and treatment of hypertrophic scar. 2> Disodium cromoglycate can inhibit slightly the proliferation and collagen synthesis of hypertrophic scar fibroblasts in vitro .
