Objective To evaluate the distribution of the GFPT2 gene polymorphismIVS2+26G>A, rs2303008, rs2303007 and rs1035411 and its association with type 2diabetes in Han people of Shanghai region of China.Methods Four common SNPs of GFPT2 gene were genotyped with PCR-RFLP andthen analyzed with transmission-disequilibrium test (TDT) and sib transmission-disequilibrium test (STDT) in 75 T2DM pedigrees (336 individuals), and haplotypescombined by the four SNPs were calculated by Genehunter software. Furthermore, theassociation of GFPT2 polymorphism with physiological and biochemical charactorswere also performed.Results (1)There were no positive results in four variants in TDT-STDTanalysis(P>0.05). (2)There were no preferential transmission of any haplotypes.(3)There were no differences between genotypes and allele genes of four SNPs,respectively.Conclusion The four variants of GFPT2 gene were not linkage with T2DM in TDT-STDT analysis, and no sighnificant association of haplotypes with T2DM and IGT wasdetected in our study. Furthermore, the studied SNPs in GFPT2 gene have no affectionon the function of pancreaticβcell and unlikely contribute to the development ofobesity.
