The special anatomic structure of pulp tissue makes it necessary to study the ability of the bacterium to cause inflammation, when we discussed the etiology and therapeutics of the pulp inflammation. Based on these, discussion of the biological change when pulp fibroblast has been invaded by P.g can give some help to curing the odontophlegmon.Objective: The purpose of this study was to investigate when pulp fibroblast had been invaded by P.g, the change of the cellular vigor and the mRNA expression level of RPP and signal transmission molecular. Methods :Establish the model of P.g viable bacteria intra-cellularly invaded pulp fibroblast in vitro. Apply the MTT to detect the effect of different density of the bacterium and different invasion time on the cellular vitality of pup fibroblast. We have used realtime-PCR to detect the mRNA expression level of NOD1 N0D2 RIP2 when P.g invaded pulp fibroblast with different density and invasion time. Results: When the density is MOI100 and MOI200, P.g can get in the pulp fibroblast, and can be cultivated when the cell was broken. Invaded by proper density (MOI100 and MOI200)of P.g, most of the pulp fibroblast can remain alive. Pulp fibroblast can express the mRNA of NOD1 NOD2 RIP2. When P.g invaded the pulp fibroblast, the transcriptional level of NOD1 N0D2 RIP2 would go up to peak at 2 hours after the invasion and then get down continually, but it still was different with control when 24 hours after the invasion. conclusion: The model of the P.g invasion the pulp fibroblast had been established sucessfully. P.g has the cell toxicity dependent on the density, while effecting the vitality of the fibroblast. Pulp fibroblast can express PRR in kytoplasm and RIP2, when P.g get in the cell, the transcription level of these molecular would up-regulate, this means pulp fibroblast can recognize bacteria in the kytoplasm, and transmit the the signal through the RIP2. when the pulp fibroblast invaded by the P.g the transcription level of NOD1 NOD2 RIP2 would change as the time pass by.
