OBJECTIVE: To investigate the inhibitory effect of compound CA-4P on human non-small cell lung cancer A549, human liver cancer cell line SMMC7721, human cervix cancer Hela cell line, and human leukaemia HL-60 cell line in vitro. And to study the effect of CA-4P on Lewis cell C57BL/6 mice transplant tumor, mice ascites cell H22, Hela cell nude mice transplant tumor, and HL-60 cell nude mice transplant tumor in vivo. METHODS: The inhibitory effect of compound CA-4P on human non-small cell lung cancer A549, human liver cancer cell line SMMC7721, human cervix cancer Hela cell line, and human leukaemia HL-60 cell line was valued by MTT analysis, cell growth curve and clone forming experiments with different concentration of CA-4P. The change of cell configuration was observed by dyeing. The inhibitory effect of compound CA-4P on Lewis cell C57BL/6 mice transplant tumor, mice ascites cell H22, Hela cell nude mice transplant tumor, and HL-60 cell nude mice transplant tumor in vivo was demonstrated by the inhibitory rate or prolongation of survival times in mice with tumour. RESULTS: After treated with CA-4P for 24 or 48 hours, MTT assay showed that the dose range from 0.625μg·ml-1 to 80μg·ml-1 had the inhibitory ratio range from 19%to 83%on A549 cell, the dose range from 0.25μg·ml-1 to 32μg·ml-1 had the inhibitory ratio range from 20%to 48%on SMMC7721 cell, the dose range from 0.5μg·ml-1 to 64μg·ml-1 had the inhibitory ratio range from 15 %to 75%and from 18%to 80%on Hela cell in 24h and 48h respectively, the dose range from 0.5μg·ml-1 to 64μg·ml-1 had the inhibitory ratio range from 15%to 79%on HL-60 cell. The average IC50 of CA-4P on A549 cell in 48h was 5.529μg·ml-1, on SMMC7721 cell in 48h was 10.294μg·ml-1, on Hela cell in 24h and 48h was 7.375μg·ml-1 and 4.294μg·ml-1 respectively, on HL-60 cell in 48h was 6.579μg·ml-1. The dose-effect relationship was described in the growth curve. The characteristic morphology typical for apoptosis was observed under microscope. The dose-effect relationship was also observed in the experimentation of Lewis cell C57BL/6 mice transplant tumor, mice ascites cell H22, and Hela cell nude mice transplant tumor in vivo by CA-4P, and the inhibitory rate or the prolongation of survival times in mice with tumour was increasingly enhanced along with the rise of CA-4P concentration. However, in the study of HL-60 cell nude mice transplant tumor in vivo we found that there were no significant effects in the least concentration of CA-4P treated group; and there were significant effects in the mid and larger concentration of CA-4P treated group. CONCLUSIONS: CA-4P could inhibit the growth of A549 cell, SMMC7721 cell, Hela cell and HL-60 cell in vitro, and its inhibition of Lewis cell C57BL/6 mice transplant tumor, mice ascites cell H22, Hela cell nude mice transplant tumor and HL-60 cell nude mice transplant tumor in vivo was distinct.
