objective to study the clinical value of NIS expression on thyroidcarcinoma diagnosis and 131I therapeutic effects prediction. Methods31 cases of thyroid carcinomas enrolled in our hospital from 1998 to 2006were included with 31 primary and 30 metastatic specimens available. Theenrollment criterion was to have thyroidectomy and resection of themetastatic carcinoma in our hospital and preoperational radiology was notpeformed. There were 19 cases of PTC, 5 FTC, 4 fvPTC, 1 IC and 2 UDTC.Using immunohistochemical method, NIS expression location, positive cellstaining and expression intensity was observed, which was calculated byimmunohistochemical scores and NIS expression level was compared betweenprimary and metastatic carcinoma. Results NIS was over-expressed onthe basolateral membrane in positive control-Grave disease tissue, and showsno staining in negative control. NIS was expressed in cytoplasm in all the 31primary carcinoma, and IHS score was over or equaled to 4 in 80.64% ofthem. Except for 2 no staining, NIS was expressed in cytoplasm in the rest 28metastatic carcinoma. NIS expression was related to the pathological type of thyroid carcinoma, strongest in PTC, then FTC, and weakest in fvPTC. NISexpression in metastatic carcinoma was related to that in primary carcinoma.Discussion NIS was over-expressed in cytoplasm in most thyroidcarcinoma, and the iodide uptaking defect was mainly due to its wronglocation. It has great potential to be applied in clinic by that it can help withthe differential diagnosis of benign and malignant thyroid diseases, especiallybetween FTA and FTC, and that it can predict the therapeutic effects of 131â… therapy following thyroid operation. Also this research has shown some lighton issues like pathologic classification of fvPTC, NIS expression in UDTC,and application of NIS expression on improving the therapeutic effects of 131â… therapy.
