Background and Objective:Primary hepatocellular caicinoma is one of the most frequent malignant tumor in our country, which attributes to the death of about 230 million people in China.The morbidity of primary hepatocellular carcinoma has been increasing in recent years. Although the early detection level and treatment technic of Primary hepatocellular caicinoma has been elevated in recent years, the survival rate of Primary hepatocellular caicinoma was not have to take on a new look. etiology unknown is one of mainnest cause among the total,whatmore metastasia and relapse occurred is the main reason of the death in the most of the patients,which have been the great obstacle to further improve survival rate of liver cancer and to overcome cancer.Etiology unknown is the main reason of the short life span of Primary hepatocellular caicinoma. hepatic cirrhosis, virus hepatitis, alcohol, adipositas, diabetes mellitus, non-Alcoholic fatty live, flavacin and other chemcial carcinogens or water and soil factor are the nanocephalous cause of hepatocarcinogenesis which has been generally accepted. In recent years someone raise a new pathogenesy, which consider that the hepatocarcinogenesis is a multitude steps and multifactorial process. chronic injury of Hepatic, immunologic response and the oxidized of DNA injury may cause gene mutation, carcinogenic gene;oncogene activated , tumor-suppressing gene devitalized, at last result in carcinogenesis.Histone deacetylase 6(HDAC6) is microtubule and HSP90 associated protein, which control genetic transcription and change cellule motor ability by deacetylased microtubule and HSP90,and then participate tumorigenesis, growth, metabasis, and the production of tumor vessel.So, it is not clear yet whether HDAC6 expression is uniform in the different part of primary hepatocellular carcinoma tissue, what is the relationship between HDAC6 expression and the factor of tumorigenesis? There are not report at present. This research detect the HDAC6 mRNA and protein expression in the primary hepatocellular carcinoma tissue and normal liver tissue,and the relationship with clinic patho- module, so we may find a new cause of hepatocarcinogenesis and metastasize to provide a new theory and strategy to cure hepatoma.Methods:The expression of HDAC6 was detected by laser capture microdissection ( LCM ) combination with reverse transcription- polymerase chain reaction(RT-PCR)and immunohistochemistry method in 36 cases of primary hepatocellular carcinoma patients primary hepatocellular carcinoma tissue and 24case normal liver tissue.Results:1.The expression of HDAC6 was detected by laser capture microdissection ( LCM ) combination with reverse transcription- polymerase chain reaction(RT-PCR) showed that The expression level of HDAC6 was higher in the primary hepatocellular carcinoma tissue than normal liver tissue.the expression level of HDAC6mRNA in the primary hepatocellular carcinoma was 0.82±0.04,and the expression value of normal liver tissue was 0.63±0.06, there was significant difference between them(t=15.81,p<0.05).The expression of HDAC6 was detected by immunohistochemistry method show that there are 27 cases were strong positive, and 9 cases were positive in primary hepatocellular carcinoma tissue;but 3 cases were strong positive and 20 cases were positive,1cases were Negative.(χ2 =25.83,P<0.05 ).2.The expression level of HDAC6 mRNA in the primary hepatocellular carcinoma tissue was related to tumor metastases, tumor encapsulstion, Edmonson grade, and AFP;whereas were not related to liver cirrhosis, ,tumor size which detected by laser capture microdissection(LCM)combination with reverse transcription-polymerase chain reaction( RT-PCR) ; this result was correspond to that of detected by immunohistochemistry method.Conclusion:HDAC6mRNA was over–expression in primary hepatocellular carcinoma tissue, which expression level was related to tumorigenesis and progress.
