Objective:To study correlation between the expression of E-cadherin and COX-2 in human gastric carcinoma and its relationship,and the clinical significance.Methods:Expression of E-cadherin and COX-2 in 63 gastric carcinoma, 56 adjacent nontumorous gastric and 15 normal gastric groups were detected by Ultrasensitive Streptavidin-Peroxidase(S-P)immunochemical staining.x~2 was used to analyze the data and the method of Spearman analyzed the correlation of E-cadherin and COX-2,there is a statistical significance when P<0.05.Results:The positive expression rate of E-cadherin in gastric carcinoma group,adjacent nontumorous gastric group and normal gastric group was 30.15%(19/63),85.71%(48/56),100%(15/15)respectively.The expression rate of E-cadherin was lower in gastric carcinoma group than that of the adjacent nontumourous gastric and the normal gastric groups(P<0.05).The expression of E-cadherin was significantly associated with lymph node metastasis,differential degree and TNM stages(P<0.05).but there was no significant correlation with gender,age,tumor location,diameter of primary tumor and the depth of invasion(P>0.05).The positive expression rate of COX-2 in gastric carcinoma group,adjacent nontumorous gastric group and normal gastric group was 73.01%(46/63),14.28%(8/46),0%(0/15)respectively.The expression rate of COX-2 was higher in gastric carcinoma group than that of the adjacent nontumourous gastric and the normal gastric groups(P<0.05).The positive expression of COX-2 was significantly associated with lymph node metastasis and TNM stages(P<0.05).but there was no significant correlation with gender, age,differential degree,tumor location,diameter of primary tumor and the depth of invasion(P>0.05).There was a negative correlation between the expression of E-cadherin and COX-2,proved by the correlation test(r_s=-0.302, P=0.016<0.05).Conclusion:The down-regulated expression of E-cadherin and the high expression of COX-2 participate invasion and diversion of gastric carcinoma together.The expression of E-cadherin and COX-2 has a close correlation. Combination of E-cadherin and COX-2 detection may be helpful to evaluate the capability of the tumor invasion and diversion of gastric carcinoma.There are indexes to know the biological behavior of gastric carcinoma and serve as fairly good markers for prognosis and therapy.
